摘 要:目的研究小檗碱对人卵巢癌细胞的抑制作用,并探讨其调控机制。方法培养人卵巢癌细胞SKOV3,给予不同浓度的小檗碱,通过CCK8法检测细胞增殖;流式细胞术检测细胞周期和凋亡,Transwell法检测细胞迁移,Western-Blot检测细胞中Cyclin Dk Bcl-2、Bax,Vimentin,E-cadhering白表达。结果小檗碱剂量依赖性地抑制卵巢癌细胞增殖;通过下调Cyclin D1表达调控细胞周期;小檗碱可明显诱导SKOV3细胞凋仁下调Bcl-2并上调Bax表达;此外,小檗碱通过调节Vimentin和E-cadherin的表达抑制细胞迁移。结论小檗碱剂量依赖性地抑制SKOV3细胞增殖和迁移,促进细胞周期停滞,诱导细胞凋亡,达到抗肿瘤效果。Objective To investigate the anti-tumor effect of berberine on human ovarian cancer cells and its regulatory mechanism.Methods Human ovarian cancer cell line SKOV3 was cultured and given berberine at different concentrations.Cell proliferation was detected by CCK8 assay,flow cytometry was used to detect cell cycle and apoptosis,Transwell method was used to detect cell migration,and Western-Blot was used to detect the protein expression of Cyclin DI,Bcl-2,Bax,Vimentin and E-cadherin.Results Berberine inhibited ovarian cancer cell proliferation in a dose-dependent manner.The cell cycle was regulated by down-regulating Cyclin DI expression.Berberine could induce apoptosis of SKOV3 cells,down-regulate Bcl-2 and up-regulate Bax expression.In addition,berberine inhibits cell migration by regulating the expression of Vimentin and E-cadherin.Conclusion Berberine can inhibit the proliferation and migration of SKOV3 cells in a dose-dependent manner,promote cell cycle arrest,induce cell apoptosis,and achieve anti-tumor effect.
