摘  要:目的明确黄芩浸提过程机理,提高黄芩提取工艺质控水平。方法基于Fick’s扩散定律,构建黄芩黄酮类单成分浸提动力学方程;采用DPPH法检测浸提不同时间点提取液的抗氧化活性;采用CCK-8法,以抑制率评价不同浸提阶段浸提液对胃癌sgc-7901细胞增殖的影响;采用脂多糖诱导小鼠RAW264.7巨噬细胞产生炎症模型,以CCK-8法和Griess法评价浸提液在不同浸提时刻的抗炎活性。结果黄芩苷浸提动力学方程为C=[11.1889×t/(M-1.5125)],在实验室条件下得到成功验证。黄芩浸提液在抗氧化、抗肿瘤和抗炎方面均具有一定的活性,长时间的加热提取对黄芩的药效并无裨益,甚至有损其抗肿瘤活性。结论揭示了黄芩浸提过程黄芩苷溶出量和工艺参数间的关系,强调把控浸提时间的重要性,为黄芩饮片提取的工艺优化和质量控制提供理论支撑。Objective To clarify the mechanism of Scutellariae Radix extraction process and improve the quality control of Scutellariae Radix extraction process.Methods Based on Fick’s diffusion law,the extraction kinetic equation of flavonoid from Scutellariae Radix was constructed;the anti-oxidant activity of extracts at different time points was detected by DPPH method;CCK-8 method was used to evaluate the effect of extracts at different extraction stages on the proliferation of gastric cancer sgc-7901 cells;a lipopolysaccharide-induced inflammation model of mouse RAW264.7 macrophages was used to evaluate the anti-inflammatory activity of the extracts at different moments of infusion by CCK-8 method and Griess method.Results The kinetic equation for the aqueous extraction of baicalin is C=[11.1889×t/(M-1.5125)],which was successfully verified under laboratory conditions.The extract of Scutellariae Radix showed anti-oxidise,anti-tumor and anti-inflammatory activities,and the excessively long heating extraction did not benefit the efficacy of Scutellariae Radix,even to the detriment of its anti-tumor activity.Conclusion The relationship between baicalin and process parameters during the extraction of Scutellariae Radix is revealed,and the importance of controlling the extraction time is emphasized,so as to provide a reference for the process optimization and quality control of extraction production of Scutellariae Radix.
