摘　　要：目的探讨长链非编码RNA(LncRNA)X染色体失活特异转录物(LncRNA XIST)在T2DM患者外周血单个核细胞(PBMC)中的表达及与胰岛β细胞功能的关系。方法收集新疆哈萨克族健康人群与T2DM患者外周静脉血行高通量测序筛选出差异性表达的LncRNA,选取新疆哈萨克族T2DM患者(T2DM组,n=30)及体检健康人群(NC组,n=30),测量身高、体重、WC、臀围、SBP、DBP,检测FPG、HbA_(1)c、FIns、TG、TC、HDL-C、LDL-C、血尿酸、BUN、血肌酐,计算BMI、WHR、胰岛素抵抗指数(HOMA-IR)、胰岛β细胞功能指数及胰岛素敏感指数;采用实时荧光定量PCR(qRT-PCR)技术检测受试者PBMC中LncRNA XIST表达水平。结果qRT-PCR检测结果显示,LncRNA XIST在T2DM组PBMC中的表达水平是NC组的3.75倍(P<0.05)。Pearson相关分析显示,新疆哈萨克族T2DM患者Lnc XIST基因表达水平与HOMA-IR呈正相关(P<0.05),与FIns呈负相关(P<0.01)。受试者工作特征(ROC)曲线显示,Lnc XIST的ROC曲线下面积为0.692(95%CI0.558~0.826,P<0.05)。结论新疆哈萨克族T2DM患者Lnc XIST基因表达水平与胰岛β细胞功能和IR有关;PBMC中LncRNA XIST表达升高可能参与新疆哈萨克族T2DM。Objective To investigate the expression of long-stranded non-coding RNA(LncRNA)X-chromosome inactivation specific transcript(XIST)in peripheral blood mononuclear cells(PBMC)of type2 diabetes mellitus(T2DM)patients and its relationship with pancreaticβ-cell function.Methods Firstly,high throughput sequencing of peripheral venous blood was performed to screen out differentially expressed LncRNA from normal and diabetic patients of Kazakh ethnicity in Xinjiang.30 T2DM patients(T2DM group)and 30 non-diabetics(NC group)were selected and accepted the measurement of height,weight,waist circumference,hip circumference,SBP,DBP,FPG,HbA_(1)c,FIns,TG,TC,HDL-C,LDL-C,SUA,BUN and Scr.BMI,WHR,insulin resistance index(HOMA-IR),isletβ-cell function index(HOMA-β)and insulin sensitivity index(HOMA-IS)were calculated.The expression levels of LncRNA XIST in PBMC of the subjects were detected by quantitative real-time fluorescence PCR(qRT-PCR)technique.Results qRT-PCR assay showed that the relative expression level of LncRNA XIST in PBMC of T2DM group was 3.75 times higher than that of NC group(P<0.05).Pearson correlation analysis showed that the expression level of Lnc XIST gene was positively correlated with HOMA-IR(P<0.05)and negatively correlated with serum insulin level(P<0.01)in Xinjiang Kazakh T2DM patients.The ROC curve showed that the area under the curve(AUC)of Lnc XIST was 0.692(95%CI0.558~0.826,P<0.05).Conclusion Lnc XIST gene expression levels in Xinjiang Kazakh T2DM patients are associated with isletβ-cell function and IR.Elevated LncRNA XIST expression in PBMC may be involved in the occurrence of T2DM in Xinjiang Kazakh population.
