摘 要:糖尿病肾病(diabetic nephropathy,DN)已成为终末期肾病的首要致病因素以及慢性肾病的主要致死原因。作为基因表达的调控因子,微小RNA(micro RNA,miRNA)及其相关的信号通路与DN的发病机制密切相关,同时也可作为肾疾病的生物预测标志物。本文从miRNA的产生与生物作用,DN的发病机制,转化生长因子β1/Smads信号通路、磷酸酰基醇3激酶/蛋白激酶B信号通路、核因子-κB信号通路、丝裂原活化蛋白激酶信号通路对DN发生发展的影响,miRNA在泛素-蛋白酶体系统中的作用,以及miRNAs作为DN的生物标志物等内容进行综述,为今后DN的诊断和治疗提供参考。Diabetic nephropathy(DN)has become the leading cause of end-stage renal disease and the main cause of death of chronic kidney disease.As regulatory factors of gene expression,microRNA(miRNA)and its related signal pathways are closely related to the pathogenesis of diabetic nephropathy,which can also be used as biological predictive markers of renal disease.This article reviews the relevant contents(production and biological effects of miRNA,the pathogenesis of DN,transforming growth factor-β1/Smads signaling pathway,phosphoacylol 3 kinase/protein kinase B signaling,nuclear factor-B signaling,mitogen-activated protein kinase signaling on DN development,the role of miRNA in the ubiquitin-proteasome system,and miRNAs as a biomarker of DN)to provide reference for the diagnosis and treatment of diabetic nephropathy in the future.
