摘 要:为了解宁夏某种鸡群孵化率及雏鸡存活率下降原因,随机采集血样进行鸡白痢、鸡伤寒抗体血清学检测,同时采集死胚及死雏肝进行病原菌分离。分离病原菌通过16S rRNA和生化试验,经多重PCR鉴定禽源沙门菌血清型并检测了14种毒力基因,测定分离株对单宁酸(TA)的最低抑菌浓度(MIC)。结果显示,鸡群鸡白痢、鸡伤寒抗体阳性率为20%;试验共分离病原菌39株,其中鸡源沙门菌29株,主要血清型为鸡白痢沙门菌26株、肠炎沙门菌2株、鼠伤寒沙门菌1株;鸡白痢沙门菌对单宁酸最为敏感,MIC为0.125~1 mg/m L。本研究揭示了该种鸡群内病原菌感染情况和携带的沙门菌毒力基因,检测了单宁酸对各分离株的MIC,为单宁酸净化沙门菌的临床应用提供了参考。To understand the reasons for the decrease in hatching rate and chick survival in a certain flock,blood samples were randomly collected for serological testing of antibodies to chicken dysentery and chicken typhoid,and dead embryos and livers of dead chicks were collected for isolation of pathogenic bacteria.The isolated pathogens were preliminarily identified by 16S rRNA and biochemical tests.Multiplex PCR was used to identify Avian Salmonella serotypes and 14 virulence genes were detected.Furthermore,and the minimum inhibitory concentration(MIC)of the isolates against tannic acid was determined.The results showed that the antibody positive rate of chicken white dysentery and chicken typhoid was 20%.39strains of pathogenic bacteria were isolated,including 29 strains of Salmonella.The main serotypes were 26 strains of Salmonella pullorum,2 strains of Salmonella enteritidis and 1 strain of Salmonella typhimurium.Salmonella pullorum was the most sensitive to tannic acid with MIC of 0.125—1.00 mg/m L.This study revealed the pathogenic bacteria infection and virulence gene carrying of Salmonella in chicken flocks.We detected the MIC of tannic acid to each isolate.It provides reference for clinical application of tannic acid in Salmonella purification.
