摘 要:目的:探究D101大孔树脂吸附法对山银花总皂苷的纯化工艺。方法:以5%香草醛-高氯酸为显色剂,利用紫外可见分光光度法测定山银花总皂苷的含量;以大孔树脂对山银花总皂苷的比吸附量和解吸率作为指标,对其吸附及解吸附性能进行研究,进而确定纯化工艺。结果:D101型大孔树脂具有较好的吸附性能,其最佳吸附工艺条件为:药液上样浓度0.2 g/m L,洗脱流速3 mL/min。上样吸附5 h后,依次用蒸馏水5 BV,30%乙醇3 BV,70%乙醇3 BV洗脱,在此条件下,获得的山银花总皂苷平均纯度为79.57%。结论:本实验确定的纯化工艺能有效分离纯化山银花总皂苷。Objective:To study the purification process of total saponins from Flos lonicerae by D101 macroporous resin adsorption.Methods:Using 5%vanillin-perchloric acid as a coloring agent,the content of total saponins of Flos lonicerae determined by UV-visible spectrophotometry,The specific adsorption amount and resolution of total mountain saponin of mountain silver flower were used as indicators to adsorb and decompose them.The adsorption performance was studied to determine the optimal purification process.Results:D101 macroporous resin has good adsorption performance.The optimal adsorption process conditions are:drug solution loading concentration of 0.2 g/mL,and elution flow rate of 3 mL/min.After the sample was adsorbed for 5 h,it was eluted with distilled water 5 BV,30%ethanol 3 BV,and 70%ethanol 3BV in this order.Under this condition,the average purity of total saponins of mountain silver flower was 79.57%.Conclusion:The purification process determined in this experiment can effectively purify total saponins of Lonicera japonica.
