摘 要:目的观察饮水铅暴露对大鼠大脑皮质、小脑、海马组织DNA连接酶Ⅲ(DNA LigⅢ)及其与氧化应激的关系。方法40只SD大鼠根据体重按随机区组法分为5组(对照组和4个暴露组),对照组自由饮用去离子水,暴露组分别饮用不同浓度的醋酸铅溶液,剂量分别为100、200、400、800 mg/L,连续染毒60 d后取大脑皮质、小脑和海马组织,Western印迹检测SD大鼠脑组织DNA LigⅢ蛋白的表达量,并测定SD大鼠血铅、脑组织铅的含量及大脑皮质、小脑和海马组织中氧化应激指标[过氧化物酶(CAT)、过氧化氢(H_(2)O_(2))、抑制OH-活力]的含量。结果染铅后,暴露组大鼠大脑皮质、小脑和海马中DNA LigⅢ蛋白表达量明显低于对照组,且随染铅剂量的升高呈逐渐下降趋势(P<0.05),并与脑组织铅、HO含量呈负相关(P<0.05),与CAT、抑制OH-活力呈正相关(P<0.05)。结论铅可通过诱导神经细胞氧化应激损伤改变脑组织DNA LigⅢ蛋白的表达水平。Objective To observe the effects of lead exposure in drinking water on the expression of DNA ligaseⅢ(DNA LigⅢ)in cerebral cortex,cerebellum and hippocampus of rats and its relationship with oxidative stress.Methods 40 SD rats were randomly divided into 5 groups(control group and 4 exposure groups)according to body weight.The control group drank deionized water freely,and the exposure group drank different concentrations of lead acetate solution,the doses were 100,200,400 and 800 mg/L.After 60 days of continuous exposure,the cerebral cortex,cerebellum and hippocampus were collected.Western blot method was used to detect the expressions of DNA LigⅢprotein in the brain tissue of SD rats.At the same time,the content of lead in blood,lead in brain and oxidative stress indicators[peroxidase enzyme(CAT),hydrogen peroxide(HO),inhibition of OH-activity]in cerebral cortex,cerebellum and hippocampus were detected.Results After lead exposure,the expressions of DNA LigⅢprotein in the cerebral cortex,cerebellum and hippocampus of rats in the exposed group were significantly lower than those in the control group,which showed a gradual decrease with the increase in lead exposure(P<0.05),and it was negatively correlated with the content of lead and HOin brain tissue(P<0.05)and positively correlated with CAT,inhibition of OH-activity(P<0.05).Conclusions Lead could change the expression level of DNA LigⅢprotein in brain tissue by inducing oxidative stress injury in nerve cells.
