海南龙血树种子超低温保存研究

摘　　要：海南龙血树是传统中药资源龙血竭的主要来源物种之一,也是国家二级重点野生保护植物。为了能够长期稳定保存海南龙血树野生种质资源,本研究采用3种超低温保存方法处理不同含水量的海南龙血树种子,通过发芽率、生理生化指标及细胞结构的综合评估,筛选出适宜海南龙血树种子超低温保存方法。结果表明,6.00%的种子含水量和直接冷冻法超低温保存对海南龙血树种子的细胞结构伤害和各项生理生化活性影响较小,且具有相对较高的发芽率(73.89%),建议将此组合作为长期保存海南龙血树种子的方法。此研究结果初步解决了海南龙血树种质资源长期保存较难的问题,为保护其野生资源提供指导意义。Dracaena cambodiana, listed as the Grade Ⅱ national key protected plant, is one of the most important species to produce resina draconis used as a traditional medicine. To find the suitable ultra-cryopreservation method to maintain the wild germplasm resources of Dracaena cambodiana under long-term storage, its seeds with different moisture contents were used as the experimental material to determine germination rate, physiological and biochemical indicators and the integrity of cytoarchitecture by three methods of ultra-cryopreservation. The results showed that the 6.00% moisture content and the direct freezing method had less effect on the cell structure damage and physiological and biochemical activities of test seeds, as well as the seeds had a relatively high germination rate of 73.89 %. After a comprehensive assessment to the test seeds, the combination of 6.00% moisture and direct freezing was identifed as the suitable cryopreservation method for long-term storage of Dracaena cambodiana seeds. The results of this study had preliminarily resolved the problem where it was hard to preserve the germplasm resources of Dracaena cambodiana for a long time, and provided guidance for the protection of its wild resources..
