摘 要:利用灵敏的超高效液相色谱-四极杆飞行时间质谱串联技术(UPLC-Q-TOF-MS)对珍稀中药资源四川阿坝野生滇黄精的80%甲醇提取液进行分析。采用电喷雾离子源(ESI)的阴离子模式对样品中的化学成分进行扫描,通过MS^(E)数据采集方式获得准分子离子峰及二级碎片等质谱信息。结合文献中已报道的黄精化学成分,建立了基于UNIFI平台的黄精数据库,通过快速筛选鉴定了包括5个黄酮类,17个甾体皂苷类等的28个化学成分,所有28个化合物为首次在四川阿坝野生滇黄精中确认,其中10个化合物为首次明确在滇黄精中发现。同时,对发现的黄酮类、皂苷类等的质谱裂解规律进行了探讨。该方法可用来快速、高效、准确评估黄精等中药活性成分。The chemical constituents from methanol extract of wild Polygonatum kingianum Coll.et Hemsl in Sichuan Aba were analyzed by ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS) technology.The pseudo-molecular ions of the chemical constituents and their fragments were detected by the negative mode of electrospray ionization(ESI) and further MS^(E) data with low and high energy level.Under the guidance of the UNIFI database and the reported mass data,twenty-eight compounds were identified,including 5 flavonoids and 17 steroidal saponins.All 28 compounds were first identified in the wild Polygonatum kingianum Coll.et Hemsl in Sichuan Aba,and 10 of them were first identified in Polygonatum kingianum Coll.et Hemsl.This method can be used to quickly assess the differences and levels of compounds in wild Polygonatum kingianum Coll.et Hemsl in Sichuan Aba and further traditional Chinese medicines,and providing an accurate scientific basis for the determination and further development of Chinese herbal medicines.
