高效液相色谱法测定甘露降糖丸中栀子苷的含量

摘　　要：目的建立高效液相色谱法测定蒙药甘露降糖丸中栀子苷的含量。方法色谱柱为SHIMADZU Shim-pack GIST C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-水(15∶85,V/V)为流动相,流速为0.8 ml/min,检测波长:238 nm,柱温:30℃,进样量:10μl。结果栀子苷在19.69~78.77μg/ml范围内线性关系良好(r=0.9998,n=14);平均加样回收率为93.87%,RSD为1.52%(n=9)。精密度和稳定性的RSD分别为0.15%和0.74%。测定2批甘露降糖丸,平均含量为5.85 mg/g,RSD为1.66%。结论该方法准确度高,专属性强,重现性好,可用于甘露降糖丸的质量控制。Objective To establish the high performance liquid chromatograghy method for the content determination of Geniposide in Mongolian medicine Ganlu Jiangtang Pills.Methods The chromatographic column was SHIMADZU Shim-pack GIST C_(18)(250 mm×4.6 mm,5μm),the mobile phase was acetonitrile-water(15∶85,V/V),the flow rate was 0.8 ml/min,detection wavelength was 238 nm,the column temperature was 30℃,and the injection volume was 10μl.Results Geniposide showed a good linearity in the range of 19.69-78.77μg/ml(r=0.9998,n=14).The average recovery was 93.87%with RSD of 1.52%(n=9).The RSD of precision and stability were 0.15%and 0.74%.The average content of 2 batches samples was 5.85 mg/g,with RSD of 1.66%.Conclusion The method is accurate,strong specificity and reproducible which can be used for the quantitation control of Ganlu Jiangtang Pills.
