摘 要:目的本研究基于ACE-AngⅡ-AT1R、ACE2-Ang(1-7)-MAS信号通路研究唐古特红景天活性部位(ACRT)抗大鼠高原缺氧性肺动脉高压的作用机制。方法将大鼠分为对照组、模型组、给药组,模型组和给药组大鼠在低压氧舱(模拟海拔4500米)内通过诱导构建高原缺氧性肺动脉高压模型。给药组分别灌胃给予250、125、62.5 mg/Kg的ACRT。给药4周后,测定大鼠肺动脉压,观察肺组织切片,测定与肺组织中ACE-AngⅡ-AT1R、ACE2-Ang(1-7)-MAS信号通路相关的蛋白表达情况。结果不同剂量给药组ACRT能显著抑制大鼠的肺动脉压升高,抑制大鼠的肺血管重塑、炎性病变,抑制大鼠的肺小动脉内皮Ⅰ、Ⅱ型上皮细胞线粒体肿胀,并调控大鼠肺组织中与ACE-AngⅡ-AT1R、ACE2-Ang(1-7)-MAS信号通路相关的蛋白活化,其中高剂量给药组效果最佳。结论ACRT具有显著的抗大鼠高原缺氧性肺动脉高压作用,其具体作用机制可能与调控ACE-AngⅡ-AT1R、ACE2-Ang(1-7)-MAS信号通路有关。Objective To investigate the mechanism of active site of Rhodiola algida anti-high altitude pulmonary hypertension in rats based on ACE-AngⅡ-AT1 R and ACE2-Ang(1-7)-MAS signaling pathways.Methods The rats were divided into three groups,including control group,model group and drug administration group.The model of high altitude pulmonary hypertension was established by induction in the hypobaric oxygen chamber(4500 meters above sea level)in the control group and drug administration group.The administration group was given 250,125 and 62.5 mg/kg ACRT by gavage respectively.The pulmonary artery pressure was measured and lung tissue sections were observed after 4 weeks of administration.The related protein expression of ACE-AngⅡ-AT1 R,ACE2-Ang(1-7)-MAS signaling pathways in lung tissues were measured as well.Results Different doses of ACRT could significantly inhibit the increase of pulmonary artery pressure,pulmonary vascular remodeling and inflammatory lesions,and the mitochondrial swelling of pulmonary arteriolar endothelial type I and II epithelial cells and regulate the activation of ACE-AngⅡ-AT1 R,ACE2-Ang(1-7)-MAS signaling pathways related proteins in rat lung tissues.Among them,the high dose group had the best effect.Conclusion ACRT has obvious anti-high altitude pulmonary hypertension in rats effect,which may be related to the regulation of ACE-AngⅡ-AT1 R,ACE2-Ang(1-7)-MAS signaling pathways.
