摘  要:目的 探讨茯苓酸(Pachymic acid,PA)对小鼠急性放射性肺损伤的防护作用机制。方法 将雄性C57BL/6J小鼠随机分为正常对照组、单纯照射组和药物干预组(包括低剂量PA+照射组、中剂量PA+照射组、高剂量PA+照射组)。采用6MV-X线(20 Gy)照射全肺,建立急性放射性肺损伤模型。药物干预组小鼠在照射前、后7 d于腹腔注射PA,正常对照组和单纯照射组小鼠给予等量橄榄油。照射14 d取材,用HE法观察小鼠肺组织病理情况;用IHC法分析p21蛋白表达情况;用RT-PCR法检测p21mRNA的基因表达水平;用ELISA法测定小鼠支气管肺泡灌洗液中的IL-6、TNF-α和TNF-β1表达水平;用SOD、MDA试剂盒检测小鼠肺组织中SOD、MDA的含量。对上述研究结果做相关分析。结果 HE结果显示,与正常对照组比,单纯照射组小鼠肺泡及肺间质增生,肺泡间隔增厚,伴有炎性细胞的浸润;与单纯照射组比,随着药物浓度的梯度式增加,小鼠肺组织炎症减轻。IHC、RT-PCR、ELISA结果显示,与正常对照组比,单纯照射组小鼠肺组织p21蛋白、p21mRNA和IL-6、TNF-α、TNF-β1表达显著升高(P<0.05);与单纯照射组比,低剂量药物干预组小鼠肺组织p21蛋白、p21mRNA和IL-6、TNF-α、TNF-β1表达差异无统计学意义(P>0.05);随着药物浓度的梯度式增加,小鼠肺组织p21蛋白、p21mRNA和IL-6、TNF-α、TNF-β1表达逐渐下降(P<0.05);与正常对照组比,单纯照射组小鼠肺组织MDA水平升高、SOD水平降低(P<0.05);与单纯照射组比,随着药物浓度的梯度式增加,小鼠肺组织SOD水平逐渐增高、MDA水平逐渐降低(P<0.05)。结论 PA可下调小鼠肺组织p21蛋白和MDA及IL-6、TNF-α、TNF-β1水平,升高SOD水平,发挥对小鼠急性放射性肺损伤的防护作用。Objective To investigate the protective mechanism of Pachymic acid(PA)against acute radiation-induced lung injury in mice.Methods Male C57 BL/6 J mice were randomly divided into normal control group,irradiation group and drug intervention group(including low dose PA+irradiation group,medium dose PA+irradiation group and high dose PA+irradiation group).The model of acute radiation-induced lung injury was established by irradiating the whole lung with 6 MV-X-ray(20 Gy).Mice in drug intervention group were intraperitoneally injected with PA before and 7 days after irradiation,while mice in normal control group and irradiation group were given the same amount of olive oil.After 14 days of irradiation,the lung tissues were collected and the pathological changes of the mice were observed by HE method.The expression of p21 protein was analyzed by IHC method.The expression level of p21 mRNA was detected by RT-PCR.The expression levels of IL-6,TNF-α and TNF-β1 in bronchoalveolar lavage fluid of mice were determined by ELISA.SOD and MDA kit were used to detect the contents of SOD and MDA in lung tissue of mice.Relevant analysis was made on the research results mentioned above.Results HE results showed that compared with the normal control group,the alveolar and interstitial hyperplasia,alveolar septum thickening and inflammatory cell infiltration were observed in the irradiation group.Compared with the irradiation group,with the gradient increase of drug concentration,the lung tissue inflammation of mice was reduced.The results of IHC,RT-PCR and ELISA showed that compared with the normal control group,the expression of p21 protein,p21 mRNA and IL-6,TNF-α and TNF-β1 in lung tissue of mice in the irradiation group were significantly increased(P0.05).With the gradient increase of drug concentration,the expression of p21 protein,p21 mRNA and IL-6,TNF-α and TNF-β1 in lung tissue of mice gradually decreased(P<0.05).Compared with normal control group,the level of MDA in lung tissue of mice in irradiation group was increased
