摘 要:目的探讨姜黄素对乳腺癌细胞生物学行为的影响及相关机制。方法人乳腺癌MCF-7细胞株分为:空白对照组、姜黄素干预A组、姜黄素干预B组、姜黄素干预C组、姜黄素干预D组;空白对照组经生理盐水处理,其余各组分别加入姜黄素进行干预,浓度分别为10μmol/L、20μmol/L、30μmol/L、40μmol/L;分别于药物作用12 h、24 h、36 h、48 h时,应用MTT实验检测细胞增殖情况,应用Transwell小室法检测细胞侵袭情况。药物作用12 h时,应用RT-PCR法检测JAK2/STAT3信号通路相关因子(p-STAT3、p-JAK2、MMP-9、MMP-2)表达情况。结果姜黄素干预组的MCF-7细胞增殖率、细胞透膜数量均明显低于空白对照组同期(P<0.05);伴随时间的进展,不同浓度姜黄素干预组的细胞增殖率明显降低(P<0.05);各时间点,姜黄素浓度越高,MCF-7细胞增殖率越低、细胞透膜数量越少(P<0.05);姜黄素干预组的MCF-7细胞中,p-STAT3、p-JAK2、MMP-9、MMP-2 mRNA表达水平均明显低于空白对照组(P<0.05)。结论姜黄素对乳腺癌细胞的增殖、侵袭具有明显的抑制作用,可能与其对JAK2/STAT3信号通路的抑制作用有关。Objective To investigate the effects and its related mechanism of curcumin on biological behavior of breast cancer cells.Methods Human breast cancer MCF-7 cell lines were divided into blank control group,curcumin intervention group A,curcumin intervention group B,curcumin intervention group C and curcumin intervention group D.In relevant experimental operations,the blank control group was treated with normal saline,and the other groups were treated with curcumin,with concentrations of 10μmol/L,20μmol/L,30μmol/L,40μmol/L,respectively.MTT assay was used to detect cell proliferation and Transwell chamber assay was used to detect cell invasion at 12h,24h,36h and 48h,respectively.RT-PCR was used to detect the expression of JAK2/STAT3 signaling pathway related factors(p-STAT3,p-JAK2,MMP-9 and MMP-2)12h after drug treatment.Results The proliferation rate and permeable membrane number of MCF-7 cells in curcumin intervention group were significantly lower than those in blank control group(P<0.05).With the progress of time,the cell proliferation rate of each group was significantly decreased(P<0.05).At each time point,the higher curcumin concentration was,the lower proliferation rate and permeable membrane number of MCF-7 cells were(P<0.05).The mRNA expression levels of p-STAT3,p-JAK2,MMP-9 and MMP-2 in MCF-7 cells in curcumin intervention group were significantly lower than those in blank control group(P<0.05).Conclusion Curcumin has significant inhibitory effect on the proliferation and invasion of breast cancer cells,which may be related to its inhibitory effect on JAK2/STAT3 signaling pathway.
