摘 要:目的:探讨丹酚酸A通过lncRNA B调控TGF-β/Smads通路改善阿霉素诱导的心力衰竭的机制。方法:细胞实验选取H2C9细胞,将其随机分为空白对照组、模型组、丹酚酸A低剂量组、丹酚酸A中剂量组、丹酚酸A高剂量组、长链非编码RNA B(lncRNA B)抑制组。采用PCR检测lncRNA B、转化生长因子-β(TGF-β)水平;采用MTT检测细胞增殖;采用流式细胞术检测细胞凋亡率;采用Western Blotting(WB)法检测TGF-β/Smads通路相关蛋白表达。动物实验选取30只雄性C57BL/6小鼠,将其随机分为对照组、阿霉素性心力衰竭模型组、丹酚酸A低浓度组、丹酚酸A中浓度组、丹酚酸A高浓度组。采用PCR检测lncRNA B、TGF-β水平;并检测各组小鼠心脏功能、生化指标水平、心肌组织病理学变化;采用WB法检测TGF-β/Smads通路相关蛋白表达。结果:细胞实验中与空白对照组相比,模型组lncRNA B和TGF-β表达较高,与模型组比,丹酚酸A各剂量组和lncRNA B抑制组表达较低,P<0.05;动物实验中与对照组相比,阿霉素性心力衰竭模型组lncRNA B和TGF-β表达较高,而经丹酚酸A干预后,表达降低,P<0.05;细胞实验中与空白对照组相比,模型组存活率较低,凋亡率较高,与模型组相比,丹酚酸A各剂量组和lncRNA B抑制组存活率较高,凋亡率较低,P<0.05;动物实验中与对照组相比,阿霉素性心力衰竭模型组左室内压最大上升速度(dp/dt_(max))、左室内压最大下降速度(-dp/dt_(max))、左心室收缩压(LVSP)水平较低,左心室舒张末压(LVEDP)水平较高,而经丹酚酸A干预后,其水平发生逆转,P<0.05;动物实验中与对照组相比,阿霉素性心力衰竭模型组生化指标较高,而经丹酚酸A干预后,生化指标水平好转,P<0.05;HE染色结果显示丹酚酸A可改善心肌组织病理损伤;细胞实验中与空白对照组相比,模型组Smad2、Smad3蛋白表达较低,TGF-β1表达升高,与模型组相比,丹酚酸A各剂量组和lncRNA B抑制组蛋白表达均�Objective:To investigate the mechanism of salvianolic acid A regulating TGF-β/Smads pathway through lncRNA B to improve adriamycin-induced heart failure.Methods:In the cell experiment,H2C9 cells were randomly divided into blank control group,model group,salvianolic acid A low-dose group,salvianolic acid A medium dose group,salvianolic acid A high-dose group and long-chain noncoding RNA B(lncRNA B)inhibition group.PCR was used to detect lncRNA B and transforming growth factor-β(TGF-β)Level;MTT was used to detect cell proliferation;The apoptosis rate was detected by flow cytometry;The expression of TGF-β/Smads pathway related proteins was detected by Western Blotting(WB)method.In the animal experiment,30 male C57BL/6 mice were randomly divided into control group,adriamycin induced heart failure model group,salvianolic acid A low concentration group,salvianolic acid A medium concentration group,salvianolic acid A high concentration group.lncRNA B and TGF-βlevels were detected by PCR.The heart function,biochemical index level and myocardial histopathological changes of mice in each group were detected.The expression of TGF-β/Smads pathway related proteins was detected by WB method.Results:In the cell experiment,compared with blank control group,the expression levels of lncRNA B and TGF-βin model group were higher,Compared to the model group,while the expression levels of salvianolic acid A each dose group and lncRNA B inhibition group were lower,P<0.05.In the animal experiment,compared with the control group,the expression levels of lncRNA B and TGF-βwere higher in adriamycin induced heart failure model group,but decreased after salvianolic acid A intervention,P<0.05.In the cell experiment,compared with blank control group,model group had lower survival rate and higher apoptosis rate.Compared with model group,salvianolic acid A each dose group and lncRNA B inhibition group had higher survival rate and lower apoptosis rate,P<0.05.In the animal experiment,compared with the control group,the levels of dp/dt_(max
