摘 要:目的研究非瑟酮(fisetin)对环磷酰胺(cyclophosphamide,CYP)导致的大鼠肝脏氧化损伤的保护作用。方法将40只清洁级健康雄性SD大鼠随机分成5组,每组8只,分别为空白组、环磷酰胺组以及非瑟酮低、中、高剂量组。除空白组外,其余各组腹腔注射环磷酰胺80 mg/kg。非瑟酮低、中、高剂量组每日50 mg/kg、100 mg/kg、200 mg/kg非瑟酮灌胃给药,连续给药14天。将SD大鼠脱颈处死后行HE染色,观察肝脏组织病理情况;采用透射电镜观察肝脏组织超微结构变化;检测血清中谷丙转氨酶(alanine aminotransferase,ALT)和谷草转氨酶(aspartate aminotransferase,AST)活性;检测肝脏超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、过氧化氢酶(catalase,CAT)活性,以及丙二醛(malondialdehyde,MDA)含量。结果与环磷酰胺组相比,非瑟酮低、中、高剂量组肝细胞变性有不同程度的改善,ALT及AST的活性剂量依赖性降低(P<0.05),肝组织抗氧化酶活性升高(P<0.05),肝组织中MDA含量降低(P<0.05)。结论非瑟酮可以抑制环磷酰胺引起的血清酶活性改变,对环磷酰胺诱导的肝脏氧化损伤具有保护作用。Objective To investigate the protective effect of fisetin on oxidative damage of liver induced by cyclophosphamide(CYP)in rats.Methods Forty male SD rats were randomly divided into 5 groups(n=8):blank group,cyclophosphamide group,low,medium and high dose of fisetin group.Except for blank group,other groups were intraperitoneally injected with cyclophosphamide 80 mg/kg.In the low,medium and high dose of fisetin group,50 mg/kg,100 mg/kg and 200 mg/kg fisetin were administered by gavage for 14 days.The liver histopathology of SD rats was observed by HE staining after decapitation;The ultrastructural changes of liver were observed by transmission electron microscope;The activities of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum were detected;The activities of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),catalase(CAT)and malondialdehyde(MDA)in liver were measured.Results Compared with cyclophosphamide group,the degree of hepatocyte degeneration was improved in varying degrees in low,medium and high dose of fisetin group,the activities of ALT and AST were decreased in a dose-dependent manner(P<0.05),but the activity of antioxidant enzymes was increased(P<0.05),the content of MDA in liver tissue was decreased(P<0.05).Conclusion Fisetin could inhibit the changes of serum enzyme activity induced by cyclophosphamide and protect the liver from oxidative damage induced by cyclophosphamide.
