摘 要:目的 评价并比较用于检测隐孢子虫和贾第鞭毛虫的2种浓缩方法,探寻最优浓缩方法。方法 按照GB/T5750.12.2006《生活饮用水标准检验方法》,利用第3方定值质控物Colorseed,制成6组12个20 L蒸馏水/水源水加标样本。富集浓缩卵囊和孢囊,浓缩分别采用离心法和抽滤法,2种浓缩方法收集到的卵囊和孢囊分别进行免疫磁分离、染色计数。结果 离心法每次最多离心4个样本且耗时大于30 min;隐孢子虫平均回收率、变异系数分别为29.67%和20.00%;贾第鞭毛虫平均回收率、变异系数分别为27.67%和15.00%。抽滤法单个样品仅需5 min;隐孢子虫平均回收率、变异系数分别为43.17%和11.00%;贾第鞭毛虫平均回收率、变异系数分别为42.00%和8.00%。结论 浓缩采用抽滤法比离心法耗时更短,回收率更高,变异系数更小。抽滤法对提高隐孢子虫和贾第鞭毛虫的检出率是有效的。Objective To evaluate and compare two concentration methods in the detection of cryptosporidium and giardia,and to explore the optimal concentration method.Methods According to Standards for Drinking Water Quality-Microbiological Parameters(GB/T 5750.12-2006),the socalled Colorseed third-party determined control materials were used,and 6 pairs of 12 20 L standard samples plus distilled/river water were prepared.The oocysts and cysts were enriched and concentrated by centrifugation and vacuum filtration method respectively,and then were magnetic separated and counted by fluorescent antibody staining.Results The centrifugation method took more than 30minutes to centrifuge 4 samples at most each time.The average recovery rate and variation coefficient for the detection of cryptosporidium was 29.67% and 20.00%,and for the detection of giardia was 27.67% and 15.00%,respectively.However,the vacuum filtration method only took 5minutes to handle a single sample.The average recovery rate and variation coefficient for the detection of cryptosporidium was 43.17% and 11.00%,and for the detection of giardia was 42.00%and 8.00%,respectively.Conclusions The vacuum filtration method takes less time than the centrifugation method,with higher recovery rate and smaller variation coefficient.The vacuum filtration method is effective to improve the detection rate of cryptosporidium and giardia.
