摘  要:【目的】从元素吸收及转运相关基因表达两方面,研究菠萝磷钾元素吸收与利用机制,为进一步开展菠萝磷和钾吸收与转运机制研究奠定基础。【方法】以菠萝叶片为供试材料,设置MS营养液、10.029 mmol/L K_(2)HPO_(4)、20.054 mmol/L KCl、1.250 mmol/L K_(2)HPO_(4)、1.250 mmol/L NaH_(2)PO_(4)5个处理,以H_(2)O(CK)为对照,测定处理后不同天数菠萝叶片的磷钾元素含量及相关转运基因的表达情况。【结果】施肥处理后,1.250 mmol/L K_(2)HPO_(4)和NaH_(2)PO_(4)处理的菠萝叶片磷元素含量显著提高,10.029 mmol/L K_(2)HPO_(4)和KCl处理的菠萝叶片磷元素含量与对照无显著差异;在施肥处理后2 d,1.250 mmol/L K_(2)HPO_(4)处理、NaH_(2)PO_(4)处理、10.029 mmol/L K_(2)HPO_(4)处理和KCl处理的菠萝叶片钾元素含量均显著提高,处理后4 d,KCl处理后菠萝叶片钾元素含量显著提高,处理后6 d,1.250 mmol/L K_(2)HPO_(4)处理菠萝叶片钾元素含量显著提高。AcPHT2、AcPHT3、AcPHT4基因在NaH_(2)PO_(4)处理后基因表达水平显著降低,AcPHT6基因的表达水平在NaH_(2)PO_(4)处理后显著升高;1.250 mmol/L K_(2)HPO_(4)处理后,AcPHT6基因的表达水平表现出先下降后升高的变化趋势,其余磷转运蛋白相关基因的表达水平均呈现先升高后下降的变化趋势,其中AcPHT1、AcPHT2、AcPHT3、AcPHT4基因的表达水平在处理后4 d达到最高。10.029 mmol/L K_(2)HPO_(4)处理后,AcNa/H2、AcNHX、AcHKT、AcKUP2基因的表达水平显著升高;AcKUP1、AcNHX基因在KCl处理后基因表达水平显著下降,AcNa/H1、AcHKT、AcKUP2基因的表达水平在KCl处理4 d后差异达到最大。【结论】不同钾源会影响菠萝对磷钾的吸收,磷钾元素间存在交互作用,影响着菠萝对磷钾元素的吸收与利用。【Objective】The mechanism of phosphorus and potassium absorption and utilization in pineapple was studied from two aspects of element absorption and transport-related gene expression,which laid a foundation for further research on the absorption and transport mechanism of pineapple phosphorus and potassium.【Method】Pineapple leaves were used as the test material,and 5 treatments were set with MS nutrient solution,10.029 mmol/L K_(2)HPO_(4),20.054 mmol/L KCl,1.250 mmol/L K_(2)HPO_(4),and 1.250 mmol/L NaH_(2)PO_(4),and H_(2)O(CK)was used as the control.The content of phosphorus and potassium in pineapple leaves on different days after treatment and the expression of related transport genes were determined.【Result】After fertilization treatment,the phosphorus content of pineapple leaves treated with 1.250 mmol/L K_(2)HPO_(4) and NaH_(2)PO_(4) was significantly increased,and the phosphorus content of pineapple leaves treated with 10.029 mmol/L K_(2)HPO_(4) and KCl had no significant difference with the control;After 2 days of treatment,1.250 mmol/L K_(2)HPO_(4) treatment,NaH_(20PO_(4) treatment,10.029 mmol/L K_(2)HPO_(4) treatment and KCl treatment all significantly increased the potassium content of pineapple leaves.After 4 days of treatment,the potassium content of pineapple leaves increased significantly after KCl treatment,and after 6 days treatment,1.250 mmol/L K_(2)HPO_(4) treatment the potassium content of pineapple leaves increased significantly.The gene expression levels of AcPHT2,AcPHT3,and AcPHT4 were significantly decreased after NaH_(2)PO_(4) treatment,and the expression level of AcPHT6 gene was significantly increased after NaH_(2)PO_(40 treatment.After 1.250 mmol/L K_(2)HPO_(4) treatment,the expression level of AcPHT6 showed a trend of first decreasing and then increasing,and the expression levels of other phosphorus transporter-related genes showed a trend of first increasing and then decreasing,among which AcPHT1,AcPHT2,AcPHT3,The expression level of AcPHT4 gene reached the highest level aft
