摘  要:【目的】垂体是动物重要的内分泌器官,生长激素细胞是垂体的重要组成细胞之一,探究生长激素细胞的分化规律对于研究垂体组织的发育以及动物发育生物学至关重要。【方法】通过体内和体外研究糖皮质激素类激素地塞米松在鹅胚垂体发育中的作用。通过向发育不同阶段的鹅胚内注射地塞米松,向体外培养的原代垂体细胞添加地塞米松,利用放射免疫法、实时荧光定量PCR法、免疫荧光染色等方法检测地塞米松对GH蛋白、GH mRNA以及GR mRNA表达变化,研究地塞米松对垂体生长激素细胞分化的影响。【结果】胚内注射的地塞米松显著提高了胚胎血浆中的GH蛋白含量,同时提高了垂体组织中的GH mRNA和GR mRNA表达量;体外培养液中添加的地塞米松提高了含有GH的垂体细胞数量,也提高了细胞中GH mRNA和GR mRNA表达量。可见,不管是蛋内注射还是体外培养液中添加地塞米松,都能有效诱导GH mRNA和蛋白在垂体细胞中表达。【结论】糖皮质激素在鹅胚垂体发育过程中具有重要作用,能促进鹅胚垂体表达GH,诱导垂体生长激素细胞的分化。【Objective】The pituitary gland is an important endocrine organ of animals, and growth hormone cells are the key part of pituitary glands. It is essential to study growth hormone cells’ developmental process for animal developmental biology. 【Method】In the present experiment, the role of glucocorticoid hormone dexamethasone was studied during the development of the embryonic pituitary gland of goose in vivo and in vitro. Dexamethasone was injected into goose embryos at different developmental stages, and dexamethasone was added into the medium of primary pituitary cells cultured in vitro. The effects of dexamethasone on GH protein, GH mRNA and GR mRNA expression were detected using radioimmunoassay, real-time fluorescent quantitative PCR and immunofluorescence staining. We could infer the relationship of dexamethasone and the differentiation of pituitary growth hormone cells during goose embryonic development. 【Result】Intraembryonic injection of dexamethasone significantly increased the level of GH protein in embryonic plasma, and increased the expression of GH mRNA and GR mRNA in pituitary tissue. The number of pituitary cells containing GH, GH mRNA and GR mRNA expression in cells were visibly improved by dexamethasone added into in vitro culture medium. In vivo and in vitro experiments have fully proved that dexamethasone could improve goose embryo pituitary cells to express GH mRNA and protein in pituitary cells.【Conclusion】It shows that glucocorticoid plays an important role in the development of goose embryonic pituitary, which can promote the expression of GH and induce the differentiation of pituitary somatotrophs in goose embryo pituitary.
