基于生信分析筛选老年性聋关键基因及相关信号通路研究

摘  要:目的利用生物信息学方法筛选老年性聋小鼠耳蜗组织中的关键基因,并分析其功能,为老年性聋的发病机制和诊治提供新思路。方法从基因表达数据库(Gene Expression Omnibus,GEO)下载GSE6045基因芯片,通过GEO2R筛选出老年性聋小鼠耳蜗组织中的差异表达基因,利用DAVID分析工具进行差异基因的基因本体论(Gene Ontology,GO)和京都基因与基因组百科全书数据库(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析,通过STRING分析差异表达基因的蛋白-蛋白相互作用(Protein-Protein Interaction,PPI),通过Cytoscape进行PPI网络的模块以及关键节点基因的分析。结果在7周龄和36周龄小鼠耳蜗组织中,共筛选出1000个差异表达基因,其中包括560个表达上调基因和440个表达下调基因。这些差异表达基因主要富集在免疫反应、中性粒细胞趋化等过程以及白细胞跨内皮迁移等通路。根据MCODE得分筛选出3个关键作用模块,基于cytoHubba中7种算法,最终筛选出Ptprc、Mmp9、Igf1这3个关键节点基因。结论本研究利用生物信息学筛选出老年性聋小鼠耳蜗组织中的差异基因及相关通路,帮助理解老年性聋的发生、发展病理机制,为寻找老年性聋新的治疗靶点提供思路。Objective To screen hub genes for presbycusis in cochlea tissues and analyze their functions using bioinformatics methods in mice to improve the understanding of the pathogenesis,diagnosis and treatment of presbycusis.Methods The GSE6045 gene chip was downloaded from the GEO database,and differentially expressed genes in cochlea tissues in mice with presbycusis were screened out by GEO2R.The DAVID online analysis tool was used to analyze the GO annotation and KEGG pathway of differentially expressed genes,and the protein-protein interaction of differentially expressed genes was analyzed by STRING.Cytoscape was used to analyze the modules and key node genes of the protein-protein interaction network.Results A total of 1000 differentially expressed genes,including 560 up-regulated genes and 440 down-regulated genes,were selected from cochlea tissues in 7-weeks-old and 36-weeks-old mice.These differentially expressed genes were mainly involved in immune responses,neutrophil chemotaxis and leukocyte transendothelial migration pathways.Three key function modules were screened out by MCODE scores,and three key node genes,Ptprc,Mmp9 and Igf1,were finally screened out based on 7 methods in cytoHubba.Conclusion In this study,bioinformatic methods were used to screen for differential genes and related pathways in cochlea tissues in mice with presbycusis to help understand the pathogenesis and development of presbycusis,and to provide basis for finding new therapeutic targets for presbycusis.
