芪贞提取物对CTX诱导小鼠免疫低下的修复作用研究

摘　　要：目的:研究芪贞提取物(QE)对环磷酰胺诱导小鼠免疫低下的修复作用。方法:ICR小鼠随机分为空白对照组、模型对照组和芪贞提取物(QE)低、中、高剂量组。除空白对照组外,其余各组均采用注射环磷酰胺(Cyclophosphamide,CTX)的方法诱发小鼠免疫低下模型,QE低、中、高剂量组灌胃不同剂量芪贞提取物(100、200、400 mg/kg),空白对照组和模型对照组灌胃同体积生理盐水。采用碳粒廓清试验、半数溶血素试验和迟发型超敏反应试验,研究芪贞提取物对小鼠的免疫修复作用。结果:与空白对照组比较,模型对照组小鼠廓清指数、吞噬指数、小鼠血清溶血素HC_(50)、脾脏指数、胸腺指数、细胞因子(IL-2、IL-4、IFN-γ)水平均降低(P<0.01),耳肿胀率升高(P<0.01);给药后,与模型对照组比较,QE各剂量组能显著改善上述指标(P<0.05,P<0.01)。结论:芪贞提取物(QE)对CTX诱导小鼠免疫低下具有明显修复作用,能提高小鼠廓清指数、吞噬指数、血清溶血素HC_(50),提高小鼠脾脏和胸腺指数,降低耳肿胀率,提高小鼠血清中细胞因子指数,其中中剂量组效果最佳。Objective:To investigate the repair effect of Qizhen extract(QE)on cyclophosphamide(CTX)-induced immunosuppression in mice.Methods:ICR mice were randomly divided into the blank control group,the model control group and the QE groups of low-dose,medium-dose and high-dose(100 mg/kg,200 mg/kg,400 mg/kg).Except for the blank control group,the other groups were injected with CTX to induce immunosuppression.The immune repair effect of QE on mice was verified by carbon clearance test,half hemolysin test,and delayed-type hypersensitivity test.Results:Compared with those in the blank control group,the K value for charcoal clearance rate,phagocytic index,serum hemolysin HC50,spleen index and thymus index,cytokines of IL-2,IL-4 and IFN-γwere decreased(P<0.01),ear swelling rate was increased in the model control group(P<0.01).After administration,compared with those in the model control group,the above indexes were all significantly improved in the QE groups(P<0.05,P<0.01).Conclusion:QE has a significant repair effect on CTX-induced immunosuppression in mice.QE can improve K value for charcoal clearance rate,phagocytic index and serum hemolysin HC50,increase spleen and thymus index,decrease ear swelling rate,and improve the serum cytokine index,among which the medium-dose group has the best effect.
