摘  要:通过荧光标记SSR基因分析技术对广东省惠东海龟国家级自然保护区2个人工繁育群体和10个疑似亲本共62只绿海龟样本进行遗传多样性和亲缘关系鉴定分析。从200个微卫星位点中筛选出20对具有多态性位点的微卫星引物;通过群体扩增实验在2个人工繁育群体中检测出78个等位基因位点,平均观测等位基因(Na)为3.9,平均观测杂合度(Ho)为0.508,平均期望杂合度(He)为0.555,平均多态性信息指数(PIC)为0.494。2个人工繁育群体的遗传多样性属于中等多态性水平。基于累积非亲排除概率来鉴定绿海龟群体和疑似亲本的亲缘关系,当可信度为95%时,2个人工繁育群体的母本鉴定率为75.00%,父本鉴定率为96.16%,双亲鉴定率为73.08%。研究结果说明,利用开发的20对微卫星引物可高效准确地应用于绿海龟亲缘关系鉴定中。本研究为建立相应的绿海龟人工繁育遗传谱系以及开展绿海龟遗传选育奠定理论基础。A total of 62 green sea turtle samples from 2 cultured populations and 10 suspected parents in National Huidong Sea Turtle Nature Reserve, Guangdong Province were analyzed for genetic diversity and genetic relationship identification by fluorescent SSR technique.From 200 microsatellite loci, 20 pairs of microsatellite primers with polymorphic loci were screened.A total of 78 alleles were detected in the two cultured populations by population amplification experiments, with an average observed allele(Na) of 3.9,an average observed heterozygosity(Ho) of 0.508,an average expected heterozygosity(He) of 0.555,and an average polymorphism information index(PIC) of 0.494.The genetic diversity of the two cultured populations was at moderate polymorphism level.Based on the cumulative non-parental exclusion probability to identify the genetic relationship between the green turtle population and the suspected parents, the identification rate of the two cultured populations was 75.00% for female parent, 96.16% for male parent, and 73.08% for both parents when the confidence level was 95%.The results indicated that the 20 pairs of microsatellite primers can be efficiently and accurately applied to green sea turtles for genetic relationship identification.This study lays the theoretical foundation for the establishment of a corresponding genetic genealogy and for genetic selection and breeding of green sea turtles in captive breeding.
