摘 要:以台湾灵芝、海南灵芝、四川灵芝和灵芝4个品种为材料,通过微波和温度组合处理、紫外线和温度组合处理,研究诱导对菌丝耐热性的影响。结果显示:4个品种对照菌丝生长停止温度分别为42℃、43℃、40℃、40℃,微波诱导的台湾灵芝和海南灵芝菌丝停止生长温度分别为43℃和44℃,紫外线诱导的四川灵芝和灵芝菌丝停止生长温度均为41℃。通过PCR分子鉴定,诱导的台湾灵芝和对照的亮带均为3条,海南灵芝的亮带比对照多2条,四川灵芝的比对照多3条,灵芝诱导和对照的均为3条。因此,本试验诱导的最佳耐热品种为海南灵芝,菌丝停止生长的温度为44℃(拟合值为43.83℃)。Ganoderma formosanum,Ganoderma hainanense,Ganoderma sichuanense and Ganoderma lucidum were used as materials to study the effects of induction on heat resistance of hyphae by combined microwave and temperature treatment and combined UV and temperature treatment.The results showed that the hyphae growth stopping temperatures of the four control cultivars were 42 ℃,43 ℃,40 ℃ and 40 ℃,respectively, while the microwave-induced mycelial growth stopping temperatures of Ganoderma formosanum and Ganoderma hainanense were 43 ℃ and 44 ℃,respectively, and the UV-induced mycelial growth stopping temperatures of both Ganoderma sichuanense and Ganoderma lucidum were 41 ℃.By PCR molecular identification, there were 3 bands both in the induced Ganoderma formosanum and the control illumination bands, 2 more bands in the induced Ganoderma hainanense than those in the control illumination bands, 3 more bands in the induced Ganoderma sichuanense than those in the control illumination bands, and 3 bands both in the induced Ganoderma lucidum and the control illumination bands.Therefore, the best heat-resistant variety induced in this experiment was Ganoderma hainanense,and the hyphae growth stopping temperature was 44 ℃(fitting value, 43.83 ℃).
