摘  要:初步探究金葡素SEC2改构蛋白2M-118对AOM-DSS诱导的结肠癌模型小鼠的药效学作用。将小鼠分为正常组(CTL)、AOM-DSS模型组(CRC)和2M-118腹腔注射给药组(2M-118),每组10只小鼠,以AOM-DSS诱导建立小鼠结肠癌模型,观察给药前后结肠癌小鼠状态变化及粪便状况,进行DAI评分;通过HE染色观察小鼠肠组织病理学变化;称取小鼠脾重及瘤重,计算免疫器官脾脏指数及抑瘤率;采用ELISA法检测小鼠结肠组织中部分炎症因子的表达情况。结果显示,AOM-DSS模型组小鼠腹泻,血便等肠炎症状明显,且后期部分小鼠出现肛外瘤情况,各项指标与正常组相比有明显变化;2M-118组腹泻、便血等肠炎症状明显减轻,与模型组相比促炎因子IL-6和TNF-α的表达量明显下降。研究结果表明,2M-118对AOM-DSS诱导的小鼠结肠癌有一定的保护作用。To initially explore the pharmacodynamics effect of Staphylococcus aureus enterotoxin SEC2 modified protein 2 M-118 on colon cancer model mice induced by AOM-DSS,C57 BL/6 male mice were randomly divided into three groups, namely, the control group(CTL),AOM-DSS model group(CRC) and 2 M-118 intraperitoneal injection group(2 M-118),with 10 mice in each group.The colon cancer in mice was induced by AOM-DSS,and the changes of colon cancer status and feces were observed before and after administration.Then disease activity index(DAI) score was performed.HE staining was used to observe the colon histopathological changes, spleen weight and tumor weight of mice were weighed, and immune organ index and tumor inhibition rate were calculated.The expression of IL-6,IL-17,TNF-α and IFN-γ were detected by ELISA.The results showed that the symptoms of diarrhea and bloody stool were obvious in the AOM-DSS model group, and some mice developed extra-anal tumors at the later stage.The indicators showed significant changes compared with the control group.The symptoms of diarrhea, hematochezia and other enteritis in 2 M-118 group were significantly alleviated.Compared with the model group, the levels of pro-inflammatory cytokines IL-6 and TNF-α significantly decreased.It was indicated that 2 M-118 had a significantly protective effect on colon cancer mice induced by AOM-DSS.
