煤气化废水驯化后硝化菌群的脱氨性能及群落分析

摘  要:试验以工业污水处理厂活性污泥为接种物,在培养装置中连续加入质量分数为10%~15%的煤气化废水曝气驯化培养,考察驯化后硝化菌液的脱氨性能,旨在获得耐受高浓度氨氮、耐酚氰的硝化菌群,同时采用16SrRNA基因文库高通量测序对其微生物群落分析。结果表明,在ρ(NH4+-N)为450 mg/L环境下,该硝化菌液较市售的2种硝化菌剂具有显著优势;经过煤气化废水驯化后的硝化菌液在pH值为8.0,温度为30℃环境条件下,NH4+-N脱除率达97%以上;经测序分析得出,驯化后样品的优势菌群主要有亚硝化单胞菌(Nitrosomonas)、硝基黄杆菌(Diaphorobacter)、假单胞菌属(Pseudomonas)。驯化后硝化菌液中有利于NH4+-N转化,促进酚、烃降解的菌群被富集,可为煤气化废水的工业化处理提供硝化菌剂。The activated sludge of industrial sewage treatment plant was used as the inoculum,and 10%~15%coal gasification wastewater was added into the continuous culture device for aeration and domestication.The ammonia removal performance of the domesticated nitrifying bacteria was investigated.The purpose was to obtain the nitrifying bacteria resistant to high concentration ammonia nitrogen and phenol cyanide.At the same time,the microbial community was analyzed by high-throughput sequencing of 16SrRNA gene library.The research results show that the nitrifying bacteria solution has significant advantages over the two commercially available nitrifying bacteria agents under the environment of ammonia nitrogen(NH4+-N)concentration of 450 mg/L;Under the environmental condition of 30℃,the removal rate of NH4+-N was more than 97%;the sequence analysis showed that the dominant flora of the samples after domestication were mainly Nitrosomonas,Diaphorobacter,Pseudomonas.After domestication,some bacteria in the nitrifying bacteria liquid that are conducive to the conversion of ammonia nitrogen and promote the degradation of phenol and hydrocarbon are enriched,and it is expected to provide nitrifying bacteria for the industrial treatment of coalgasification wastewater.
