摘　　要：建立白虎汤中芒果苷、知母皂苷BⅡ、甘草苷、甘草酸单铵盐共4种主要药效成分的高效液相色谱测定方法。采用Agilent ZORBAX SB-Aq色谱柱(4.6 mm×250.0 mm,5μm);芒果苷溶于稀乙醇,以乙腈-0.2%冰醋酸溶液(15∶85)为流动相,检测波长258 nm;知母皂苷BⅡ溶于30%丙酮溶液,流动相选择乙腈-水(25∶75),柱温30℃,流速1.0 mL/min,DAD-ELSD检测器漂移管温度70℃,载气流速2.0 L/min;甘草苷、甘草酸单铵盐溶于70%乙醇,以乙腈-0.05%磷酸溶液为流动相,梯度洗脱,检测波长237 nm。结果表明白虎汤提取液(1 g/mL)中芒果苷、知母皂苷BⅡ、甘草苷、甘草酸单铵盐质量浓度分别为1.079 mg/mL、4.270 mg/mL、0.490 mg/mL、0.790 mg/mL。该研究建立了稳定可靠的高效液相色谱检测方法,可为白虎汤相关制剂的质量控制研究与应用提供科学参考。A high-performance liquid chromatography method was established for the determination of four main medicinal components in Baihutang,including mangiferin,timosaponin BⅡ,glycyrrhizin and ammonium glycyrrhizinate.An Agilent ZORBAX SB-Aq column(4.6 mm×250.0 mm,5μm)was used for HPLC analysis.Mangiferin was dissolved in dilute ethanol,using acetonitrile-0.2%glacial acetic(15∶85)as the mobile phase,UV detection wavelength was 258 nm;timosaponin BⅡwas dissolved in 30%acetone,and acetonitrile-water(25∶75)as the mobile phase.The chromatographic analysis were performed at 30℃with flow rate of 1.0 mL·min-1,and the detector was DAD-ELSD detector drift tube temperature 70℃,carrier as flow rate 2.0 L·min-1.Glycyrrhizin and ammonium glycyrrhizinate were dissolved in 70%ethanol,acetonitrile-0.05%phosphoric acid solution was used as the mobile phase,gradient elution was carried out,and the detection wavelength was 237 nm.The content of mangiferin,timosaponin BⅡ,glycyrrhizin and ammonium glycyrrhizinate in Baihutang was 1.079 mg·mL-1,4.270 mg·mL-1,0.490 mg·mL-1and 0.790 mg·mL-1,respectively.In this study,a stable and reliable HPLC detection methodis established,which can provided a scientific reference for the quality control research and application of Baihutang related preparations.
