摘 要:目的采用“谱-效”相关研究筛选米泔水炮制后射干发挥抑制耐甲氧西林金黄色葡萄球菌作用的药效物质基础。方法通过回流水提取、回流20%乙醇提、回流40%乙醇提、回流60%乙醇提四种不同提取方法得到米泔水炮制射干提取液,分别建立其UPLC指纹图谱。以耐甲氧西林金黄色葡萄球菌为研究对象,采用琼脂打孔法进行体外抑菌研究,测定抑菌圈大小。采用灰色关联方法分析米泔水炮制射干提取液体外抑菌结果与高效液相指纹图谱共有峰相关性,建立射干“谱-效”关系。结果四种不同方式提取的药液在体外抑菌实验中,生药量1g/ml的药液能够很好的抑制耐甲氧西林金黄色葡萄球菌(MRSA)。四种不同提取方式的射干UPLC指纹图谱共确定有29个共有蜂,其中有9个已知成分。通过灰色关联谱效分析结果显示14个共有蜂对药效起主要作用。结论建立射干米泔水炮制品对体外耐甲氧西林金黄色葡萄球菌的谱-效关系,为射干逆转多重耐药菌的耐药性药效物质基础研究提供参考,为射干质量标志物的建立提供依据。Objective Screening the pharmacodynamic substance basis of inhibiting methicillin-resistant Staphylococcus aureus by Shegan(Belamcanda chinensis)Processing with Rice-washed Water used Spectral-effect correlation.Methods Shegan(Belamcanda chinensis)Processing with Rice-washed Water were prepared by reflux water extraction,20%reflux alcohol ex⁃traction,40%reflux alcohol extraction and 60%reflux alcohol extraction,and established the UPLC fingerprints respectively.To study methicillin-resistant Staphylococcus aureus(MRSA),Using AGAR punching bacteriostasis experiment in vitro bacteriosta⁃sis circle,determination of bacteriostatic ring size.And using grey relational analysis to analyze the correlation between the bacte⁃riostatic efficacy and HPLC fingerprint in Shegan(Belamcanda chinensis)Processing with Rice-washed Water,establish the Spectrum-activity of shegan.Results In vitro bacteriostatic experiments,the extracts of four different methods showed that the crude drug dosage of 1g/mL could inhibit methicillin-resistant Staphylococcus aureus(MRSA)well.A total of 29 common wasps were identified by UPLC fingerprints of four different extraction methods,including 9 known components.The results of grey correlation spectrum analysis showed that 14 common wasps played an important role in the efficacy.Conclusion To establish the spectrum-effect relationship between Shegan(Belamcanda chinensis)Processing with Rice-washed Water and methicillin-re⁃sistant Staphylococcus aureus in vitro,to provide reference for the basic research on the pharmacodynamic substances for the re⁃versal of drug resistance of multiple drug resistant bacteria,and to provide basis for the establishment of markers for the quality of Shegan.
