摘  要:Knoevenagel反应是一种经典的缩合反应,常用于增加分子的共轭以构建具有给体-受体(D-A)结构的化合物,而此类偶极分子往往对环境的粘度变化有响应。基于此考虑,该文提出了一种综合性的实验教学方案,合成了一个对粘度敏感的荧光探针PySQ,筛选出了最佳的反应条件,并测试了其紫外-可见吸收光谱和荧光光谱,验证了探针对粘度的高灵敏响应性能。该实验包含有机物的合成、提纯和性能测试,将有机化学与分析化学紧密结合,且所得探针在生物成像领域颇具潜力,因此可以很好地调动学生的积极性,提高实验教学的效果。Knoevenagel reaction is a classical condensation reaction, which is often used to expend the π-conjugation of a molecule and generate the donor-acceptor(D-A) type compound. Interestingly, such dipolar D-A molecules are commonly responsive to environmental viscosity change. Based on this consideration, a comprehensive experimental scheme for teaching is proposed in this paper. A viscosity-sensitive fluorescent probe PySQ is prepared, the optimum reaction conditions are selected and then the UV-vis absorption spectra and fluorescence spectra are tested to verify its high sensitivity toward viscosity. This experiment includes the synthesis,purification and property characterization of an organic compounds. It closely integrates organic chemistry with analytical chemistry, and the obtained probe holds great promise in biological imaging, which are supposed to mobilize students’ enthusiasm and improve the effect of experimental teaching.
