摘 要:结合临床实际检测需求和免疫标记领域研究热点,设计了基于量子点阳离子交换反应检测金葡菌A型肠毒素的综合性实验,用于医学专业病原生物学与免疫学实验教学。利用量子点修饰金葡菌A型肠毒素单克隆抗体,制备了单克隆抗体-量子点荧光标记物,并优化了基于阳离子交换反应的量子点信号检测体系,最终应用于金葡菌A型肠毒素免疫学检测方法的建立。实验结果显示,所建立的基于量子点阳离子交换反应的金葡菌A型肠毒素免疫学检测法灵敏度高、特异性好。该综合性实验将相关理论原理与技术创新相结合,加深了学生对免疫标记与检测技术教学内容的理解,提高了专业操作技能,激发了科研兴趣。Combined with the actual clinical detection needs and research hotspots in the field of immunolabeling,a comprehensive experiment based on cation exchange reaction in quantum dots to detect staphylococcus aureus type A enterotoxin(SEA) was designed for the experimental teaching of pathogenic biology and immunology in medical specialty. The monoclonal antibody quantum dots fluorescent conjugate was prepared by modifying the monoclonal antibody of SEA with quantum dots. The quantum dots signal detection system based on cation exchange reaction was optimized and used in the immunological detection method of SEA. The results showed that this immunological detection method had high sensitivity and specificity. This comprehensive experiment which combines relevant theoretical principles with technical innovation can deepen students’ understanding of the teaching content of immune labeling and detection technology, improve professional operation skills and stimulate their interest in scientific research.
