摘 要:【目的】探究不同日龄家兔血清生殖激素及相关激素受体基因在家兔生长发育过程中对精子生成及对睾丸功能的调节作用,为家兔初情期内分泌等生殖生理研究提供理论依据。【方法】采用酶联免疫法和荧光定量PCR法,分别对家兔血清生殖激素FSH、LH和T的含量及其受体基因FSHR、LHR和AR的表达状况进行研究分析。【结果】随家兔日龄的增加,血清中FSH含量在45日龄达到最高,且30、45、105日龄组均极显著高于其他各组(P<0.01);LH和T含量在150日龄均达到最高,且LH含量在30、120、150日龄组均极显著高于45、60日龄组(P<0.01);T含量在135、150日龄组均显著或极显著高于其他各日龄组(P<0.05或P<0.01)。此外,60日龄组FSHR相对表达量显著高于120、150日龄组(P<0.05),30、60、150日龄组LHR相对表达量均显著高于90日龄组(P<0.05),而AR相对表达量随日龄的增加呈递减趋势(P>0.05)。【结论】公兔初情期启动时,需要较高浓度的LH及高浓度T的作用,但FSH处在较低的水平;公兔性成熟前血清中FSH、LH浓度变化与相应受体表达趋势一致,但AR呈弱表达。【Objective】In order to provide theoretical basis for the reproductive physiology research such as endocrine mechanism of the initiation of early estrus in rabbits,the effect of serum reproductive hor‐mones and related hormone receptor genes on spermatogenesis and the regulation of testicular function dur‐ing the growth and development of rabbits at different ages were investigated.【Method】The content of se‐rum reproductive hormones and the expression of their receptor genes in rabbits were determined by enzyme-linked immunosorbent assay(ELISA)and fluorescence quantitative PCR.【Result】With the in‐crease of rabbit age,the serum FSH content reached the highest at the age of 45 days,and the serum FSH content of the groups at the ages of 30,45 and 105 days was significantly higher than that in other groups(P<0.01).The contents of LH and T were the highest at 150 days of age,and those in 30,120 and 150 days age groups were significantly higher than those in 45 and 60 days age groups(P<0.01).T contents in 135-and 150 days age groups were significantly higher than that in other groups(P<0.05 or P<0.01).In addition,the relative expression level of FSHR in 60 days age group was significantly higher than that in 120 and 150 days age group(P<0.05),the relative expression level of LHR in 30,60 and 150 days age group was significantly higher than that in 90 days age group(P<0.05),and the relative expression level of AR decreased with the increase of age(P>0.05).【Conclusion】The high concentration of LH and T but lower FSH were required for the initial estrus of female rabbit.Furthermore,the changes of serum FSH and LH concentrations in male rabbits before sexual maturity were consistent with the expression trend of the corresponding receptors,while AR was weakly expressed.
