摘  要:目的:探究miR-129-5p在糖尿病视网膜病变(DR)中的作用及调控机制。方法:采用实时荧光定量PCR检测miR-129-5p在人视网膜微血管内皮细胞模拟体内高糖环境中和构建的糖尿病大鼠中的表达。进行功能实验EdU和CCK8的测定以探索miR-129-5p在视网膜血管功能障碍中的作用。通过Matrigel管形成实验验证视网膜血管功能障碍中的作用。结果:在糖尿病老鼠模型和模拟高糖环境下miR-129-5p表达降低(P<0.01),糖尿病条件下miR-129-5p的上调抑制了人视网膜微血管内皮细胞的增殖、迁移和Matrigel管形成(P<0.01)。结论:miR-129-5p抑制DR的增殖和Matrigel管的形成,为DR提供了潜在的治疗策略。Objective:To investigate the role and regulatory mechanism of miR-129-5p in diabetic retinopathy.Methods:Real-time fluorescent quantitative PCR was used to detect the expression of miR-129-5p in human retinal microvascular endothelial cells in a simulated high glucose environment and in diabetic mice model developed.Functional experiment using EdU and CCK8 was performed to explore the role of miR-129-5p in retinal vascular dysfunction,of which was verified by Matrigel tube formation assay.Results:The expression of miR-129-5p was decreased in diabetic mouse models and simulated high glucose environment(P<0.01),and up-regulation of miR-129-5p under diabetic condition inhibited the proliferation,migration and Matrigel tube formation of human retinal microvascular endothelial cells(P<0.01).Conclusion:miR-129-5p can inhibit the proliferation of diabetic retinopathy and the formation of Matrigel tubes.The findings may provide a potential treatment strategy for diabetic retinopathy.
