清热解毒化浊片对内毒素性肝损伤大鼠肠道sIgA、pIgR的影响

摘　　要：目的:观察清热解毒化浊片对内毒素性肝损伤大鼠肠黏膜免疫球蛋白A(sIgA)、多聚免疫球蛋白合体(pIgR)的影响。方法:以高脂饮食加白酒灌胃法建立内毒素性肝损伤大鼠模型,将造模成功的18只大鼠随机分为模型组、治疗组、阳性对照组,并设正常组,每组各6只。药物干预4周后观察各组大鼠肝功能[丙氨酸氨基转移酶(AST)、天冬氨酸氨基转移酶(ALT)],sIgA、pIgR水平,以及肝脏组织病理。结果:与模型组比较,治疗组AST、ALT均下降(P<0.05),但较正常组高(P<0.05),sIgA、pIgR均升高(P<0.01或P<0.05),但较正常组低(P<0.01或P<0.05);与模型组相比,治疗组肝脏组织肝细胞脂肪变及炎性细胞浸润程度均有改善。结论:清热解毒化浊片能改善内毒素性肝损伤大鼠的肝功能,减轻其炎性细胞浸润及肝细胞脂肪变程度,作用机制与减少肠黏膜sIgA、pIgR的表达、保护肠黏膜免疫屏障有关。Objective:To investigate the effect of Qingre Jiedu Huazhuo tablets on secretory immunoglobulin A(sIgA)and polymeric immunoglobulin receptor(pIgR)in the intestinal mucosa of rats with endotoxic hepatic injury.Methods:A rat model of endotoxic hepatic injury was established by high-fat diet and liquor by gavage,and after successful modeling,18 rats were randomly divided into model group,treatment group,and positive control group,with 6 rats in each group;6 normal rats were selected as normal group.After 4 weeks of drug intervention,the rats were observed in terms of liver function[aspartate aminotransferase(AST)and alanine aminotransferase(ALT)],levels of sIgA and pIgR,and liver histopathology.Results:Compared with the model group,the treatment group had significant reductions in AST and ALT(P<0.05),but with significantly higher levels than the normal group(P<0.05),and the treatment group had significant increases in the levels of sIgA and pIgR(P<0.01 or P<0.05),but with significantly lower levels than the normal group(P<0.01 or P<0.05).Compared with the model group,the treatment group had improvements in hepatocyte steatosis and inflammatory cell infiltration in liver tissue.Conclusion:Qingre Jiedu Huazhuo tablets can improve the liver function of rats with endotoxic hepatic injury and alleviate the degree of inflammatory cell infiltration and hepatocyte steatosis,possibly by reducing the expression of sIgA and pIgR in intestinal mucosa and protecting the immunological barrier of intestinal mucosa.
