摘 要:为调查牛诺如病毒(BNoV)在新疆不同地区牛场中的感染情况,本实验针对BNoV的RdRp基因设计特异性引物,采用RT-PCR方法对新疆乌鲁木齐、塔城、伊犁、喀什4个地区的7个牛场共230份牛粪便样品进行检测,并获得塔城地区一株BNoV开放阅读框1(ORF1)基因,对其进行遗传进化、同源性及基因重组分析。流行病学调查结果显示,新疆地区BNoV阳性率为17.83%(41/230),其中塔城地区阳性率最高(60.00%,9/15)。对检测的BNoV ORF1基因进行遗传进化分析,结果显示,其属于GIII.2型BNoV,与中国四川地区OK032546.1病毒株相似性最高(94.6%),聚为一支;同源性分析结果显示,本研究中检测到的两株BNoV的ORF1基因序列之间的同源性为100%,均与OK032546.1病毒株相应基因序列同源性最高(96.6%)。ORF1编码的氨基酸序列分析显示两株BNoV ORF1与MZ573179.1病毒株同源性最高(98.35%),且与国内外其他GIII.2型病毒株同源性在76.1%以上。通过对ORF1基因重组分析,结果显示,本研究检测到的BNoV与国内外经典株ORF1基因无基因重组现象。本研究填补了新疆地区对BNoV基因序列研究的空白,为BNoV感染的防制提供了参考依据。To investigate the infection of bovine norovirus(BNoV)in cattle farms in different regions of Xinjiang,specific primers targeting RdRp gene were designed for detection of the BNoV.Reverse transcription-polymerase chain reaction(RT-PCR)technology was used in this study.A total of 230 cattle feces samples from 7 cattle farms in 4 regions of Xinjiang Urumqi,Tacheng,Yili and Kashgar were tested,and an open reading frame 1(ORF1)gene from BNoV was obtained in Tacheng region.Genetic evolution,gene homology and genetic recombination analysis were performed.The epidemiological survey results showed that the positive rate of BNoV in Xinjiang was 17.83%(41/230),of which Tacheng had the highest positive rate(60.00%,9/15).The phylogenetic tree analysis of the obtained BNoV ORF1 gene showed that it belongs to the GIII.2 type BNoV ORF1,which has the highest similarity(94.6%)with the OK032546.1 virus strain in Sichuan,China,and the two viruses clustered into one branch.The homology analysis showed that the BNoV ORF1 gene of these two viruses shares 100%in the nucleotide sequence,with the highest nucleotide homology(96.6%)with the OK032546.1 strain and share the highest amino acid homology with the MZ573179.1 strain(98.35%).The homology of other GⅢ.2 virus strains at home and abroad is above 76.1%.The recombination analysis showed no genetic recombination phenomenon occurred in ORF1 gene between the BNoV obtained in this study and the classic virus strains at home and abroad.The results of this study fill in the blank of BNoV gene sequence research in Xinjiang,and provide a reference for BNoV control.
