摘 要:目的 探讨桂皮醛对高血压小鼠血压和血管功能的影响及其机制。方法 选择SPF级C57BL/6雄性小鼠30只,随机分成为对照组、模型组和药物组,每组各10只小鼠。通过血管紧张素Ⅱ(AngⅡ)灌注构建高血压小鼠模型,其中模型组和药物组小鼠均按750 ng/(kg·min)输注AngⅡ,药物组小鼠腹腔注射50 mg/kg桂皮醛,对照组小鼠注射等量生理盐水。灌注28 d后,测量小鼠收缩压和舒张压。采用苏木素-伊红(HE)、弹力蛋白(VVG)和马松(Masson)染色,评估血管形态和胶原沉积情况。检测小鼠血清白细胞介素(IL)-1、IL-6、肿瘤坏死因子(TNF)-α和细胞间黏附分子(ICAM)-1水平。流式细胞术检测主动脉上T淋巴细胞与巨噬细胞的表达。结果 与对照组比较,模型组小鼠的收缩压和舒张压均明显升高(P<0.05);与模型组比较,药物组小鼠的收缩压和舒张压均显著降低(P<0.05)。病理染色结果显示:对照组小鼠主动脉结构完整,未见明显病理改变;模型组小鼠主动脉增厚,血管周围胶原沉积,而药物组小鼠主动脉厚度和胶原含量均显著低于模型组(P<0.05)。与对照组比较,模型组小鼠血清IL-1、IL-6、TNF-α和ICAM-1含量显著升高,差异有统计学意义(P<0.05);而与模型组相比,药物组小鼠血清IL-1、IL-6、TNF-α和ICAM-1含量明显降低,差异有统计学意义(P<0.05)。与对照组比较,模型组小鼠主动脉T淋巴细胞和巨噬细胞表达显著升高,差异有统计学意义(P<0.05);而与模型组相比,药物组小鼠主动脉T淋巴细胞和巨噬细胞表达明显降低,差异有统计学意义(P<0.05)。结论 桂皮醛可以降低高血压小鼠的血压,抑制血管重塑和炎症反应。Objective To investigate the role and mechanism of cinnamaldehyde on blood pressure and vascular function in hypertensive mice. Methods Thirty male C57BL/6 mice were randomly divided into four groups: control group, model group, and drug group. The model of hypertension in mice was established by continuous infusion with angiotensin Ⅱ(AngⅡ). The model group and drug mice were infused with Ang Ⅱ at a dose of 750ng/(kg·min). The drug group mice were intraperitoneally injected with cinnamaldehyde at a 50mg/kg dose, and the control group mice were injected with the same amount of normal saline. After 28 days, systolic pressure and diastolic pressure were measured.Hematoxylin-eosin(HE), Van Gieson(VVG), and Masson staining evaluated the vascular morphology and collagen deposition. The serum levels of IL-1, IL-6, tumor necrosis factor(TNF)-α, and intercellular cell adhesion molecule(ICAM)-1 were detected. The expression of T lymphocytes and macrophages in aorta was detected by flow cytometry.Results Compared with the control group, the vasodilation response was increased, while the vasoconstriction response was decreased in the model group(P<0.05). Compared with the model group, the vasodilation response was increased,while the vasoconstriction response was decreased in the drug group(P<0.05). The HE, VVG, and Masson staining results showed that the wall of the aorta in the model group was significantly thickened, and the collagen content was increased,while the aortic thickness and collagen content in the drug group was increased significantly lower than those in the model group(P<0.05). Compared with the control group, the expression of IL-1, IL-6, TNF-α, and ICAM-1 in the serum of mice in the model group was significantly increased(P<0.05);while compared with the model group, the expression of IL-1, IL-6, TNF-α, and ICAM-1 in the serum of mice in the drug group was significantly decreased(P<0.05).Compared with the control group, the expression of T lymphocytes and macrophages in the model group was signifi
