摘  要:改进硫酸铵法纯化单克隆抗体,优化硫酸铵的饱和度,让最终饱和度达45%,通过两次沉淀,使单克隆抗体的纯度进一步提升,再利用醋酸缓冲液去除杂蛋白,最终得到便于保存的单克隆抗体。优化后的硫酸铵法提取出蛋白质的纯度大大提高,且浓度和效价都高于传统方法,能纯化出与亲和层析纯度相媲美的单克隆抗体。优化后的硫酸铵沉淀法简单便捷,易于操作且纯化抗体纯度高,也便于抗体保存。To improve the purification of monoclonal antibody by ammonium sulfate method,the saturation of ammonium sulfate was optimized so that the final saturation reached 45%,and the purity of the monoclonal antibody was further improved through two precipitations,and then the acetic acid buffer was used to remove impurities,and finally a monoclonal antibody that was easy to store was obtained.The purity of the protein extracted by the optimized ammonium sulfate method is greatly improved,and the concentration and titer are higher than those of the traditional method,and the monoclonal antibody comparable to the purity of affinity chromatography can be purified.The optimized ammonium sulfate precipitation method is simple and convenient,easy to operate,and the purified antibody has high purity,which is convenient for antibody storage.
