摘 要:【目的】分析凝结芽孢杆菌蛋雏鸡肠道菌群的变化。【方法】选取1日龄蛋雏鸡288只,分为对照组和凝结芽孢杆菌组,每组72只。饲养28 d后,每组选取6只蛋雏鸡,收集盲肠内容物,提取样品DNA,采用MiSeq PE300高通量测序平台进行测序分析。【结果】共获得121.8万条高质量序列,基于相似度大于97%的原则聚类后共获得1926个OTU(Operational Taxonomic Unit)。与对照组相比,凝结芽孢杆菌低剂量组、凝结芽孢杆菌中剂量组和凝结芽孢杆菌高剂量组的4种α多样性指数存在差异显著(P<0.05)。在属水平上,添加凝结芽孢杆菌低剂量组、凝结芽孢杆菌中剂量组和凝结芽孢杆菌高剂量组的另枝菌属、毛螺菌属和巴恩斯氏菌属相对丰度均显著高于对照组(P<0.05),而拟杆菌属、布劳特氏菌属、梭菌属和乳杆菌属的相对丰度均显著低于对照组(P<0.05)。凝结芽孢杆菌高剂量组与相应的对照组相比,有1个菌目、6个菌科和18个菌属有显著差异。差异菌群的功能主要集中在氨基酸运输与代谢、碳水化合物运输与代谢及能量的产生和转换等相关基因上。凝结芽孢杆菌高剂量组与对照组相比差异显著的通路有11条,主要是苯丙氨酸、泛酸盐代谢和辅酶合成等通路。【结论】凝结芽孢杆菌通过酪氨酸、色氨酸的生物合成以及烟酰胺代谢和辅酶合成等信号通路,增加有益菌的相对丰度,维持菌群的相对动态平衡,来调节机体的肠道健康。[Objective]Analysis of changes in intestinal flora of Bacillus coagulans in laying chicks.[Methods]A total of 288 one-day-old laying chickens were selected.They were divided into control group and Bacillus coagulans group,72 in each group.After 28 days of feeding,6 chickens in each group were selected for collecting cecal contents and extracting sample DNA.Sequencing analysis was performed using MiSeq PE300 high-throughput sequencing platform.[Results]A total of 1.218 million high-quality sequences were obtained,and a total of 1926 OTUs(translated into Chinese)were obtained after clustering based on the principle of similarity>97%.Compared with the control group,the diversity indices of 4αin low dose,medium dose and high dose Bacillus coagulans groups were significantly different(P<0.05).At the genus level,the relative abundances of Cladella,Spironella and Barnella in low dose,medium dose and high dose groups of Bacillus coagulans were significantly higher than those in control group(P<0.05),and the relative abundances of Bacteroidetes,Brucella,Clostridium and Lactobacillus were significantly lower than those of control group(P<0.05).There were significant differences in 1 order,6 families and 18 genera of Bacillus coagulans in the high-dose group compared with the corresponding control group.The function of the differential flora mainly focused on amino acid transport and metabolism,carbohydrate transport and metabolism,energy production and conversion and other related genes.Compared with the control group,11 pathways were significantly different in the high-dose group of Bacillus coagulans,including biosynthesis of phenylalanine,synthesis of pantothenate and coenzyme,etc.[Conclusion]Bacillus coagulans regulates the intestinal health of the body by increasing the relative abundance of beneficial bacteria and maintaining the relative dynamic balance of the flora through the biosynthesis of tyrosine and tryptophan,as well as nicotinamide metabolism and coenzyme synthesis.
