摘  要:目的 研究ω-3鱼油脂肪乳剂(ω-3FOFE)通过醛类应激激活抗氧化通路,探讨其对大鼠肺缺血再灌注的保护作用。方法 选择24只SD雄性大鼠,随机分成4组(n=6):NS5组(生理盐水注射5 d)、FO5组(ω-3FOFE注射5 d)、FO3组(ω-3FOFE注射3 d)、FO1组(ω-3FOFE注射1 d)。确定ω-3FOFE预处理通过醛类应激激活抗氧化信号通路时间。实验结束取大鼠肺组织,检测肺组织丙二醛(MDA)、谷胱甘肽(GSH)含量及Nrf2和HO-1抗氧化信号通路蛋白表达。再另外选择18只大鼠,随机分成3组(n=6):Sham组(假手术组)、I/R组(缺血再灌注组)、FO组(ω-3FOFE预处理模型+缺血再灌注组)。测定各组肺组织湿干重比(W/D)、肺组织病理改变、细胞凋亡情况、GSH含量、Nrf2和HO-1蛋白表达。结果 与NS5组、FO3组及FO1组相比,FO5组的MDA含量、GSH含量、Nrf2和HO-1蛋白表达均显著增加(P<0.05)。与Sham组相比,I/R组肺组织W/D值明显升高(P<0.05),肺泡结构明显破坏,细胞凋亡显著增加,肺组织GSH含量、Nrf2和HO-1蛋白表达均显著下降(P<0.05);与I/R组相比,FO组肺组织W/D值明显下降(P<0.05),肺泡结构显著改善,细胞凋亡显著降低;肺组织GSH含量、Nrf2和HO-1蛋白表达均显著增加(P<0.05)。结论 ω-3FOFE通过醛类应激激活抗氧化信号通路从而减轻了肺缺血再灌注损伤。Objective To investigate the protective effect of ω-3 fish oil fat emulsion(ω-3 FOFE) on pulmonary ischemia/reperfusion(I/R) injury by activating the antioxidant pathway through aldehyde stress.Methods Totally 24 SD male rats were selected and randomly divided into 4 groups(n=6):NS5 group(5 days of normal saline injection),FO5 group(5 days of ω-3 FOFE injection),FO3 group(3 days of ω-3 FOFE injection),FO1 group(1 day of ω-3 FOFE injection).The time of activating antioxidant signaling pathway through aldehyde stress by ω-3 FOFE pretreatment was determined.At the end of the experiment,the lung tissues of the rats were collected to detect the contents of MDA and GSH and the expression of Nrf2 and HO-1 proteins.Another 18 rats were selected and divided into 3 groups(n=6):Sham group(sham operation group),I/R group(ischemia-reperfusion group),FO group(ω-3 FOFE+I/R group).The lung tissues of the rats were collected to measure the wet-to-dry weight ratio(W/D),observe the alveolar structure and detect the apoptosis,GSH contents,and expression of Nrf2 and HO-1.Results Compared with NS5 group,FO3 group and FO1 group,the contents of MDA and GSH and the expression of Nrf2 and HO-1 proteins in FO5 group were significantly increased(P<0.05).Compared with Sham group,the W/D value in I/R group was significantly increased(P<0.05),the alveolar structure was significantly damaged,and apoptosis was significantly increased;the content of GSH and the expression of Nrf2 and HO-1 proteins were significantly decreased(P<0.05).Compared with I/R group,the W/D value in FO group was significantly decreased(P<0.05),the alveolar structure was significantly improved,and apoptosis was significantly decreased;the content of GSH and the expression of Nrf2 and HO-1 proteins were significantly increased(P<0.05).Conclusion ω-3 FOFE attenuates pulmonary I/R injury by activating antioxidant signaling pathways through aldehyde stress.
