摘 要:目的探讨妊娠期小鼠砷(As)暴露对孕中期胎鼠脑组织全基因组mRNA表达的影响。方法将受孕第0天(D0)小鼠分为As暴露组和正常对照组,分别经口给予亚砷酸钠75μg·L^(-1)水溶液或纯净水,在受孕D12处死孕鼠,采集胎鼠脑组织,采用illuminaHiSeq高通量测序平台对全基因进行转录组测序,用GO和KEGG富集分析方法对2组间有差异表达的基因mRNA进行功能及信号通路富集分析。结果As暴露组胎鼠脑组织中有165个基因表达上调,998个表达下调;差异基因在功能上集中富集于神经突触形成及神经递质传递功能相关区域,在信号通路上主要富集于与神经发育、细胞凋亡和氧化应激等相关的通路。结论妊娠期As暴露可引起胎鼠脑组织mRNA表达异常,从而影响神经系统正常发育。OBJETIVE To investigate the effect of arsenic(As)exposure during pregnancy on whole genome mRNA expression in brain tissue of fetal mice in the second trimester.METHODS Newly pregnant mice were divided into the As exposed group and normal control group.They were fed with sodium arsenite(AaAsO_(2))aqueous solution 75μg·L^(-1)and purified water respectively.The pregnant mice were killed on the 12^(th) day of pregnancy(D12).The brain tissue of fetal mice was collected.The whole gene transcriptome was sequenced by RNA transcriptome sequencing(RNA-seq)using the Illu⁃mina HiSeq high-throughput sequencing platform,and the function and signal pathway of differentially expressed gene mRNA between the two groups were enriched and analyzed using GO and KEGG enrich⁃ment analysis methods.RESULTS A total of 165 genes were up-regulated and 998 genes were down-regulated in arsenic exposed brain tissue.The differential genes were concentrated not only in areas where neurites were formed and neurotransmitters worked but also in signal pathways related to nerve development,apoptosis and oxidative stress.CONCLUSION Arsenic exposure during pregnancy can cause abnormal mRNA expression during the development of the fetal nervous system,thus affecting the normal development of the nervous system.
