摘  要:目的 探讨小青龙汤合止痉散加减对幼龄哮喘豚鼠肺组织炎症及血清IL-17、组胺、LXA4、LTs水平的影响。方法 将72只幼龄雄性豚鼠随机分为正常组(Normal组)、模型组(Model组)、中药高剂量干预组(CMHDI组)、中药中剂量干预组(CMMDI组)、中药低剂量干预组(CMLDI组)、地塞米松组(Dex组),HE染色观察豚鼠一般形态和肺组织病理学情况,ELISA方法检测血清脂氧素A4(LXA4)、白三烯(LTs)、白介素-17(IL-17)和组胺水平,Western blot方法检测肺组织E-selectin与AMCase的表达。结果 经过药物治疗后,豚鼠一般形态学和病理组织均有改善。与Model组相比较,药物干预组血清LXA4、LTs、IL-17和组胺水平均显著降低(P<0.05),其中CMHDI组豚鼠降低最为明显(P<0.001);与Model组相比较,药物干预组豚鼠肺组织中E-selectin、 AMCase蛋白表达水平均降低(P<0.05),其中CMHDI组豚鼠降低最为明显(P<0.001)。结论 小青龙汤合止痉散加减能够下调幼龄哮喘豚鼠血清中LXA4、LTs与组胺水平以及肺组织Eselectin、AMCase蛋白的表达。Objective To investigate the effects of addition and subtraction of xiaoqinglong decoction and antispasmodic powder on lung inflammation and serum IL-17, histamine, LXA4 and LTs levels in young asthmatic guinea pigs. Methods Seventy-two young male guinea pigs were randomly divided into normal group(Normal group), the model group(Model group), chinese medicine high dose intervention group(CMHDI group), chinese medicine middle dose intervention group(CMMDI group), chinese medicine low dose intervention group(CMLDI group), and the dexamethasone group(Dex group). The general morphology and lung histopathology of guinea pigs were observed by HE. ELISA was used to detect lipoxygenin A4(LXA4), leukotriene(LTs), interleukin-17(IL-17) and histamine levels in serum. The expression of E-selectin and AMCase were detected by Western blot. Results After drug treatment, the general morphology and pathological tissue of guinea pigs were improved. Compared with model group, levels of LXA4, LTs, IL-17 and histamine in serum of drug intervention group were significantly decreased(P<0.05), especially in CMHDI group(P <0.001). Compared with model group, the expression of E-selectin and AMCase in lung tissues of guinea pigs in drug intervention group were decreased(P<0.05), and the decrease was most significant in CMHDI group(P<0.001). Conclusion Addition and subtraction of xiaoqinglong decoction and antispasmodic powder can down-regulate levels of LXA4, LTs and histamine in serum and the expression of E-selectin and AMCase in lung tissues of young asthmatic guinea pigs.
