摘  要:目的 探讨右美托咪定在大鼠脑缺血再灌注损伤的作用及可能机制。方法 采用线栓法制备大鼠脑缺血再灌注模型,随机将大鼠分为假手术组(Sham组)、模型组(MCAO/R组)和右美托咪定处理组(Dex组);于术后24h对各组大鼠进行神经功能评分和组织病理学的检测;采用TUNEL染色法检测脑细胞的凋亡;采用ELISA检测IL-1β、IL-6、TNF-α、IL-10的水平;Western blot方法检测PI3K和p-Akt的表达。结果 与MCAO/R组相比,DEX能够显著提高脑缺血再灌注大鼠的神经功能评分,改善组织病理学损伤,显著降低脑细胞的凋亡,显著抑制IL-1β、IL-6、TNF-α的表达水平,促进IL-10的释放,显著上调脑缺血再灌注大鼠脑组织PI3K和p-Akt等蛋白的表达(P<0.05)。结论 右美托咪定改善脑缺血再灌注损伤与激活PI3K/Akt信号通路相关。Objective To explore the effect and possible mechanism of dexmedetomidine in cerebral ischemiareperfusion injury of rats. Methods Middle cerebral artery occlusion(MCAO) was established by suture method. The rats were randomly divided into Sham group, MCAO/R group and Dex group. The neurological function score and histopathological examination were performed 24 hours after operation. TUNEL staining was used to detect the apoptosis of brain cells. The level of IL-1β, IL-6, TNF-α and IL-10 was detected by ELISA. The expression of PI3K and p-Akt was detected by Western blot. Results Compared with the MCAO/R group, DEX significantly improved the neurological function score and histopathological damage, reduced brain cell apoptosis, inhibited the expression of IL-1β, IL-6 and TNF-α, and up-regulated the expression of IL-10, significantly upregulated the expression of PI3K and p-Akt in brain tissue of rats with cerebral ischemia-reperfusion(P<0.05). Conclusion DEX could improve the injury after cerebral ischemia-reperfusion via activating PI3K/Akt signaling pathway.
