摘 要:目的 研究姜黄素对TNF-α诱导的HFLS-RA细胞增殖、凋亡、侵袭、迁移的影响及其与Wnt/β-catenin蛋白的关系。方法HFLS-RA细胞分为对照组、诱导组、姜黄素5μmol/L组和姜黄素10μmol/L组。MTT实验检测各组细胞增殖能力;流式细胞术检测各组细胞凋亡情况;Transwell实验和划痕实验检测细胞侵袭迁移能力;Western blot检测细胞中Wnt/β-catenin及TNF-α蛋白表达水平。结果 与对照组相比,诱导组和姜黄素组细胞增殖、侵袭与转移能力显著增强,细胞凋亡数减少,Wnt、β-catenin和TNF-α蛋白表达明显增加;与诱导组相比,姜黄素组细胞增殖、侵袭与转移能力显著降低,细胞凋亡数显著增加,Wnt、β-catenin和TNF-α蛋白表达显著下降。结论 姜黄素抑制TNF-α诱导的HFLS-RA细胞增殖、侵袭与迁移,并诱导其凋亡,其机制可能与调控Wnt/β-catenin蛋白有关。Objective To study the effects of curcumin on proliferation, apoptosis, invasion and migration of TNF-α-induced HFLS-RA cells and its relationship with Wnt/β-catenin protein. Methods HFLS-RA cells were cultured and divided into control group, induction group, curcumin 5μmol/L group and curcumin 10μmol/L group. Cell proliferation was measured by MTT assay. Cell apoptosis was detected by flow cytometry. Transwell assay and scratch assay were used to detect the invasion and migration of cells. The expression of Wnt/β-catenin and TNF-α were detected by Western blot. Results Compared with the control group, the proliferation, invasion and migraition of cells in the induced group and curcumin group were significantly enhanced, the number of apoptosis cells was decreased, and the expressions of Wnt, β-catenin and TNF-α were significantly increased. Compared with the induction group, the proliferation, invasion and metastasis of cells in curcumin group were significantly decreased, the number of apoptosis cells was significantly increased, and the expression of Wnt, β-catenin and TNF-α were significantly decreased. Conclusion Curcumin inhibits the proliferation, invasion and migration of HFLS-RA cells induced by TNF-α, and induces apoptosis, which may be related to the regulation of Wnt/β-catenin protein.
