摘 要:本研究优化了高效液相色谱法检测戊二醛含量的实验条件,建立软式内镜表面戊二醛消毒剂残留量的检测方法。超纯水采集内镜表面残留的戊二醛消毒剂,采用2,4-二硝基苯肼(DNPH)为衍生剂,70%乙腈-30%磷酸盐缓冲液(pH2.5)为流动相,C18色谱柱分离后在360 nm波长处测定戊二醛的含量。戊二醛与DNPH在50%的乙腈酸性水溶液中40℃反应20 min,形成E,E和E,Z两种异构体。戊二醛的浓度和两种异构体的峰面积和成正比,标准曲线线性范围为0.1~20.0 mg·L^(-1);检出限为0.03 mg·L^(-1);加标回收率93.7%~98.3%,精密度(RSD)小于3%。建立的分析方法能够准确、灵敏的测定内镜表面戊二醛的残留量,样品加标回收率和精密度良好,适用于医院软式内镜表面戊二醛残留的测定。This study aimed to optimize the experimental conditions of detecting glutaraldehyde content by high performance liquid chromatography(HPLC),and establish a method for detecting residual glutaraldehyde disinfectant on the surface of flexible endoscope.The residual glutaraldehyde disinfectant on the surface of endoscope was extracted with ultra-pure water.2,4-dinitrophenylhydrazine(DNPH)was used as a derivative,70%acetonitrile-30%phosphate buffer(pH2.5)was used as mobile phase,and the content of glutaraldehyde was determined at 360 nm after separation on a C18 column.Glutaraldehyde reacted with DNPH in 50%acetonitrile aqueous solution at 40 °C for 20 min to form E,E and E,Z isomers.The concentration of glutaraldehyde was proportional to the peak area of the two isomers,and the linear range of the standard curve was 0.1~20.0 mg·L^(-1) .The detection limit was 0.03 mg·L^(-1);the recoveries of spiked samples were 93.7%~98.3%,and the precision was less than 3%.The established analytical method can accurately and sensitively determine the glutaraldehyde residual on the surface of endoscope.The recovery rate and precision of sample addition are good,which is suitable for the determination of glutaraldehyde residual on the surface of flexible endoscope in hospitals.
