摘  要:目的七氟烷代谢产物六氟异丙醇(HFIP)有部分以游离形式存在体内,有潜在的麻醉效能。文章旨在使用2.32%浓度的七氟烷麻醉大鼠,检测麻醉不同时长后血中游离HFIP的生成情况,并探究其可能对苏醒及运动协调能力恢复时间的影响。方法选择8~10周龄雄性SD大鼠52只进行运动协调能力训练,采用随机数字表法将通过训练的48只大鼠分成3组(0.5 h组、1 h组、1.5 h组),每组16只。3组大鼠分别接受2.32%浓度的七氟烷麻醉0.5 h、1.0 h及1.5 h,每组内随机选取8只大鼠行腹主动脉采血检测游离HFIP含量,另外8只大鼠用于麻醉后苏醒期的观察。以翻正反射恢复作为麻醉后苏醒的指标,以通过转棒实验作为麻醉后运动协调能力恢复的指标,观察麻醉后苏醒时间和运动协调能力恢复时间。结果麻醉1 h组大鼠体内游离HFIP含量(0.081%±0.008%)明显高于麻醉0.5 h组(0.063%±0.007%)及麻醉1.5 h组(0.060%±0.007%),差异均有统计学意义(P<0.01);0.5 h组大鼠麻醉后苏醒时间[(5.0±0.5)min]分别长于1.0 h组[(3.4±0.6)min]及1.5 h组[(2.9±1.0)min],差异均有统计学意义(P<0.01),但1 h组和1.5 h组两组间比较,大鼠麻醉后苏醒时间无统计学意义(P>0.05)。3组大鼠麻醉后运动协调能力恢复时间差异无统计学意义(P>0.05)。结论在1.5 h内使用2.32%的七氟烷其代谢产物游离HFIP对大鼠苏醒期无明显影响。Objective The sevoflurane metabolite hexafluor isopropanol(HFIP)partially exists in the body in free form and has potential anesthetic efficacy.This paper aims to detect the generation of free HFIP in blood of rats anesthetized with 2.32%sevoflurane,and to explore its possible effects on the recovery time of recovery and motor coordination ability.Methods Fifty-two male SD rats aged 8-10 weeks were selected for motor coordination training,and 48 rats were divided into 3 groups(0.5 h group,1 h group and 1.5 h group)by random number table method,with 16 rats in each group.The rats in the three groups were anesthetized with 2.32%sevoflurane for 0.5 h,1.0 h and 1.5 h,respectively.Eight rats in each group were randomly selected for abdominal aortic blood sampling to detect the content of free HFIP,and the other eight rats were used to observe the recovery period after anesthesia.The recovery of righting reflex was used as the index of recovery of motor coordination ability after anesthesia,and the recovery time of recovery of motor coordination ability after anesthesia was observed by rod turning test.Results The content of free HFIP in 1 h anesthesia group(0.081%±0.008%)was significantly higher than that in 0.5 h anesthesia group(0.063%±0.007%)and 1.5 h anesthesia group(0.060%±0.007%),and the differences were statistically significant(P<0.01).The recovery time of rats after anesthesia in 0.5 h group[(5.0±0.5)min]was longer than that in 1.0 h group[(3.4±0.6)min]and 1.5 h group[(2.9±1.0)min],and the differences were statistically significant(P<0.01).However,there was no significant difference in the recovery time of rats after anesthesia between 1 h group and 1.5 h group(P>0.05).There was no significant difference in recovery time of motor coordination ability among the three groups after anesthesia(P>0.05).Conclusion The use of 2.32%sevoflurane metabolite free HFIP within 1.5 h had no significant effect on the recovery period of rats.
