摘　　要：掌叶覆盆子(Rubus chingii Hu)是重要的药用植物,由于基因组数据缺乏,导致其分子生物学研究进展相对缓慢。本研究采用根尖染色体制片技术来确定其倍性;利用流式细胞术和K-mer分析方法对其基因组大小进行了测定。结果如下,掌叶覆盆子根尖染色体数目为2n=2x=14;采用流式细胞术,以萝卜‘Saxa’为内标测定的掌叶覆盆子基因组大小为(255.39±7.58)Mb;K-mer分析估测的掌叶覆盆子基因组大小为262.2 Mb,杂合率为0.91%,重复序列比例为42.54%;根据各分析指标,推测该掌叶覆盆子基因组为一般复杂基因组。本研究结果将为掌叶覆盆子的全基因组测序研究提供重要参考。Rubus chingii Hu is a traditional Chinese herb,which has a very high medical value.Owing to the lack of its genomic resource,the development and application of molecular biology of R.chingii is still limited.In this study,we identified the ploidy of R.chingii by root-tip squashing methods and estimated the genome size of R.chingii by flow cytometry and K-mer analysis.The main findings are as follows:the number of chromosomes in the root tip of R.chingii was 2n=2x=14;According to the results of flow cytometry,the genome size of R.chingii was(255.39±7.58)Mb;K-mer analysis estimated that the genome size of R.chingiis was about 262.2 Mb,the hybridity rate was about 0.91%,and the proportion of repeated sequences was about 42.54%;The genome of R.chingiis was classified as a common complex genome.Accordingly,the results of this study provide an important database reference for the follow-up work of whole-genome sequencing.
