摘 要:为研究青海枸杞果实不同发育时期抗坏血酸代谢的变化,以枸杞‘柴杞一号’果实为试验材料,采用紫外分光光度法分析抗坏血酸含量、相关酶活性,采用荧光定量RCR技术测定基因表达变化。结果表明:枸杞果实在不同发育期,ASA含量、酶活性、基因表达的变化趋势不同。ASA含量在M2期(绿果期,花后14~19 d)最高,GLDH活性在M4期(黄熟期,花后25~30 d)最高,AO活性在M3(变色期,花后20~25 d)、M4期最高,APX活性在M5期(红熟期,花后34~40 d)最高,MDHAR活性M3期最高,DHAR活性在M5期最高,GR活性在M4期最高。GLDH和GR酶活性呈显著正相关关系(P<0.05),代谢酶基因表达与对应酶活性变化趋势相似。研究结果补充了枸杞果实发育时期ASA累积的研究,为枸杞果实抗坏血酸的代谢机理研究提供线索。In order to study the changes of ascorbic acid metabolism in different developmental stages of Lycium chinese,the fruit of Lycium chinese’Chaiqi1’was used as experimental material.The ascorbic acid(ASA)content and related enzyme activities were analyzed by ultraviolet spectrophotometry,and the gene expression was determined by fluorescence quantitative RCR.The results showed that the changes of ASA content,enzyme activity and gene expression were different in different developmental stages of Lycium chinese fruit.ASA content was the highest in M2 stage(green fruit stage,14~19 days after anthesis),GLDH activity was the highest in M4 stage(yellow maturity stage,25~30 days after anthesis),AO activity was the highest in M3(discoloration stage,20~25 days after anthesis)and M4 stage,APX activity was the highest in M5 stage(red maturity stage,34~40 days after anthesis),MDHAR activity was the highest in M3 stage,DHAR activity was the highest in M5 stage,GR activity was the highest in M4 stage.There was a significant positive correlation between GLDH and GR enzyme activity,and the change trend of metabolic enzyme gene expression and enzyme activity was similar.The results supplement the research on ASA accumulation during fruit development of Lycium chinese,and provide clues for the study of ascorbic acid metabolism mechanism of Lycium chinese fruit.
