摘 要:目的:借助同位素标记相对与绝对定量(i TRAQ)技术筛选针刺督脉治疗阿尔茨海默病(AD)蛋白靶点,以期为研究针刺治疗阿尔茨海默病的机制提供新思路。方法:将8只7月龄APPswe/PS1dE9转基因雄性幼鼠随机分为模型组和针刺组,每组4只。适应性饲养1周后,针刺督脉治疗组小鼠每只每天在清醒状态下针灸督脉百会、风府、神庭与水沟穴1次,行捻转手法,留针20 min,持续28 d后,采用Morris水迷宫和旷场实验检测各组小鼠学习、记忆和运动能力。行为学实验结束后,采用i TRAQ技术检测各组小鼠海马蛋白质组差异表达,并采用Western blot法对各组小鼠海马组织中FAM98A、SLC3A1及SLC17A7蛋白表达进行验证。结果:与模型组比较,针刺督脉治疗后小鼠第3、4天潜伏期缩短,穿越平台次数增多,中心运动距离和中心区域运动距离增加;且与模型组相比,针刺督脉治疗后小鼠的海马组织中FAM98A、SLC3A1及SLC17A7蛋白表达水平显著上升(P<0.05)。结论:针刺督脉治疗能够改善AD小鼠的学习认知能力,其治疗机制与FAM98A、SLC3A1及SLC17A7蛋白表达改变相关。Objective:To screen the protein targets of needling DU meridian in treatment of Alzheimer s disease(AD)by using i TRAQ technology,with a view to provide new ideas of the mechanism of acupuncture treatment for AD.Methods:Eight 7-month APPswe/PS1dE9 transgenic mice were randomly divided into the model group and the acupuncture group,with 4 mice in each group.After 1 week of adaptive feeding,the acupuncture group was treated with needling Baihui(DU20),Fengfu(DU16),Shenting(DU24)and Shuigou(DU26),once a day under awake state.The needles were retained for 20 minutes and the treatment duration was 28 days.Morris water maze and open field experiments were used to test the learning,memory and exercise abilities of mice in each group.After the behavioral experiment,i TRAQ technology was used to detect the differential expression of the hippocampal proteome of the mice,and Western blot was used to verify the protein expressions of FAM98A,SLC3A1 and SLC17A7 in the hippocampus.Results:Compared with those in the model group,the incubation period of mice on the 3rd and 4th day was shortened,the number of crossing the platform was increased,the movement distance of the central area was increased;and the protein expressions of FAM98A,SLC3A1 and SLC17A7 were increased significantly in the acupuncture group(P<0.05).Conclusion:Needling DU meridian can improve the learning and cognitive ability of AD mice;its mechanism may be related to the protein expressions of FAM98A,SLC3A1 and SLC17A7.
