摘  要:目的探讨陈皮苷对乙醇诱导的胃溃疡小鼠保护作用及其相关机制。方法应用乙醇(0.2 ml/20 g)诱导BALB/c小鼠建立胃溃疡模型,检测小鼠血清与胃组织中超氧化歧化酶(SOD)的活性、丙二醛(MDA)及胃内髓过氧化物酶(MPO)含量;检测小鼠血清与胃组织白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)水平。同时,采用PAS染色对小鼠胃组织进行病理学评价,采用蛋白印迹法检测小鼠胃组织中MAPK/NF-κB信号通路蛋白表达。结果陈皮苷可明显降低模型组小鼠胃组织的MPO水平,升高血清和组织中的SOD水平,降低血清和组织中的MDA水平,降低血清和组织中的IL-1β,IL-6,TNF-α炎症因子水平(P<0.01)。病理结果显示,陈皮苷能够减轻胃组织的黏膜下水肿,使白细胞浸润抑制和溃疡面积减少;同时,蛋白印迹结果显示,陈皮苷还能明显降低胃组织P-ERK/ERK、P-JNK/JNK、P-P38/P38、P-P65/P65及P-IkBa/IkBa水平(P<0.01)。结论陈皮苷对乙醇诱导小鼠胃溃疡具有胃保护作用,其机制可能与抑制MAPK/NF-κB信号通路有关。Objective To reveal the mechanism of hesperidin against ethanol-induced gastric ulcer in mice.Methods Ethanol(0.2 ml/20 g)was used to induce gastric ulcer in mice.The activity of superoxide dismutase(SOD)and level of malondialdehyde(MDA)instomachs and serum,and content of myeloperoxidase(MPO)instomachs of mice were detected,and the levels of interleukin-6(IL-6),interleukin-1β(IL-1β),and tumor necrosis factor-α(TNF-α)in serum and stomachs of mice were measured.Periodic Acid Schiff(PAS)stainingwas employed to evaluate the pathological changes of mouse stomach.The protein expression of mitogen-activated protein kinase(MAPK)/nuclear factor-kappa B(NF-κB)signaling pathway in mouse stomachwas detected by Western blotting.Results Hesperidinreduced MPO level in stomach,increased the SOD level in serum and stomach,and down-regulated the levels of MDA,IL-1β,IL-6,and TNF-αin serum and stomach of gastric ulcer mice(P<0.01).In addition,it alleviated submucosal edema of gastric tissue,inhibited leukocyte infiltration,and reduced ulcer area.Moreover,it down-regulated the levels of phosphorylatedextracellular signal-related kinase(P-ERK)/ERK,phosphorylatedc-Jun N-terminal kinase(P-JNK)/JNK,phosphorylated p38(P-P38)/P38,phosphorylatedp65(P-P65)/P65,and phosphorylatedIkBa(P-IkBa)/IkBa in gastric tissue(P<0.01).Conclusion Hesperidin is effective for the ethanol-induced gastric ulcer in mice,and the mechanism is the likelihood that it inhibits the MAPK/NF-κB signaling pathway in mice.
