摘  要:成束状阿拉伯半乳糖蛋白(FLAs)在木质部发育过程中起重要调控作用,对山新杨(Populus davidiana×P.alba var.pyramidalis)FLA基因进行生物信息学及其在山新杨不同组织中的表达模式进行了分析,筛选与木质部发育相关的FLA基因,为鉴定其调控山新杨材性形成的调控机制及分子改良奠定基础。利用拟南芥FLA家族蛋白序列在山新杨基因组序列中进行比对,共获得11条PdbFLAs,编码区长度为540至1 440 bp。对其蛋白理化性质、高级结构、系统进化、基序分布、启动子元件等进行分析。结果表明:蛋白结构和基序分析可将PdbFLAs分为3个亚类,每个亚类的蛋白结构和基序组成较为保守;系统进化分析可将PdbFLAs分为4组,这其中PdbFLA1、PdbFLA2、PdbFLA4、PdbFLA9与毛果杨、拟南芥中调控本质部发育相关基因聚为一组,推测该组内的PdbFLAs也具有类似功能;在PdbFLAs启动子中发现了与光、脱落酸(ABA)、胁迫响应和次生壁形成相关的重要顺式元件。对人工弯曲处理的山新杨木质部表型进行分析,发现树干出现偏心生长,应拉木组织被染成绿色,显示其纤维素质量分数高于对应木和直立木,木质化程度有所降低。利用荧光定量RT-PCR对PdbFLAs在根、茎、叶以及应拉木、对应木和直立木不同组织的表达模式进行分析,结果显示:PdbFLA1在茎和应拉木中高度表达,说明在山新杨木质部发育过程中PdbFLA1具有重要的调控作用。Fasciclin-like arabinogalactan proteins(FLAs) play an important role in the development of xylem. The bioinformatics and expression patterns analysis of 11 FLA genes in the different tissues of Populus davidiana×P. bolleanawere performed to screen the FLA gene involved in xylem development of P. davidiana×P. bolleana and lay the foundation for further research on the regulationalmechanism of FLA gene and molecular improvement of wood properties of P. davidiana×P. bolleana. The Arabidopsis FLA family protein sequence were used to blast with the P. davidiana×P. bolleana genome data, 11 PdbFLAs were cloned and the length of the coding region was from 540 to 1 440 bp. Their protein physical and chemical properties, advanced structure, phylogenetic tree, conserved motifs and cis-acting elements were analyzed. According to the protein structure and motif PdbFLAs can be divide into 3 subtypes, and each subtype are relatively conservative. Phylogenetic analysis can divide PdbFLAs into 4 groups. Group Aincluds PdbFLA1/2/4/9, which may be related to xylem development. Important cis-acting elements related to light, abscisic acid(ABA), stress response and secondary wall formation were found in the PdbFLAs promoters. The phenotype of the poplar xylem under the artificially bending was analyzed, the trunk grew eccentrically, and the tension wood tissue was dyed green, indicating that its cellulose content was higher than that of the opposite wood and normal wood, and the degree of lignification was reduced. The expression patterns of PdbFLAs in different tissues including roots, stems, leaves, and tension wood, opposite wood and normal wood were analyzed using RT-PCR. The results show that PdbFLA1 is highly expressed in stems and tension wood, indicating that PdbFLA1 plays an important regulatory role in the development of the xylem of Populus davidiana×P. bolleana.
