摘 要:目的:探讨蟛蜞菊水煎液对慢性阻塞性肺疾病(COPD)模型大鼠血清、肺组织、清肿瘤坏死因子α(TNF-α)、白细胞介素8(IL-8)、白细胞介素6(IL-6)、超氧化物歧化酶(SOD)、谷胱甘肽转移酶(GSH-Px)含量的影响。方法:雄性SD大鼠分为5组:空白组(n=7)、模型组(n=7)、蟛蜞菊高剂量组(6.3 g/kg,n=7)、蟛蜞菊中剂量组(3.15 g/kg,n=6)、蟛蜞菊低剂量组(1.575 g/kg,n=7);采用脂多糖(LPS)气管滴注联合烟熏制备COPD大鼠模型,历时11周。造模第8周开始干预,直至造模结束,共4周;干预结束取材,苏榛-伊红(HE)染色观察气管、肺组织病理形态变化,检测肺组织、TNF-α、IL-6、IL-8含量,SOD、GSH-Px活力。结果:与模型组大鼠比较,蟛蜞菊各剂量组大鼠气管黏膜结构破坏及肺气肿病变程度明显改善,肺组织、血清中高水平的IL-6、IL-8、TNF-α均不同程度降低;SOD、GSH-Px活力不同程度升高。其中,蟛蜞菊高剂量组大鼠降低肺组织、血清TNF-α、IL-6、IL-8效果最显著,蟛蜞菊中剂量组提高SOD、GSH-Px活力效果最显著,与模型组大鼠比较,差异均有统计学意义(P<0.01)。结论:蟛蜞菊水煎液能够改善COPD模型大鼠肺气肿样病变,其机理可能与抗炎、提高抗氧化酶活力有关。Objective:To investigate the effects of Pengqiju Decoction on TNF-α,IL-8,IL-6,SOD and GSH-Px in serum and lung tissues of rats with COPD.Methods:Male SD rats were randomly divided into 5 groups:blank group(n=7),model group(n=7),and Pengqiju Decoction high-dose group(PQJ,6.3 g/kg,n=7),medium-dose group(PQJ,3.15 g/kg,n=6),and low-dose group(PQJ,1.575 g/kg,n=7).Lipopolysaccharide(LPS)tracheal drip and fumigation were combined to make COPD rat model and lasted for 11 weeks in total.The treatment intervention was began at 8 w after modeling,and continued for 4 weeks.The samples were taken at the end of the intervention,and then the pathological morphology of trachea and lung was observed by HE stain.The contents of interleukin-6(IL-6),interleukin-8(IL-8),tumor necrosis factor-α(TNF-α),superoxide dismutase(SOD),and glutathione transferase(GSH Px)in serum and lung tissue were detected.Results:Compared with the model group,the rats in three treatment groups showed significant improvement in trachea integrity and emphysema lesion,a decrease in the contents of TNF-α,IL-6 and IL-8 in varying degrees,and an increase in the activities of SOD and GSH-Px both in lung tissue and serum.In particular,the high-dose group of Pengqiju Decoction had the most significant effect on reducing the contents of TNF-α,IL-6 and IL-8 in lung tissue and serum(P<0.01).Conclusion:Pengqiju Decoction can improve emphysema-like lesionsin COPD model rats,and the mechanism may be ralated to anti-inflammation and antioxidant enzymes activity.
