摘 要:目的:分析北豆根转录组信息特征。方法:采用二代高通量测序技术对北豆根进行转录组测序,并进行系统的生物信息学分析。结果:转录组测序共得到37346个Unigene,平均长度986 nt,其中有20562个Unigene得到注释。采用微卫星识别工具(MISA)方法共鉴定出81种重复单元共9948个简单重复序列(SSRs)。挖掘到21个Unigene参与北豆根活性成分的生物合成,涉及(S)-乌药碱N-甲基转移酶、多酚氧化酶、酪氨酸转氨酶、伯胺氧化酶、天冬氨酸转氨酶(细胞质)、天冬氨酸转氨酶(线粒体)、天冬氨酸转氨酶(叶绿体)和酪氨酸脱羧酶等8种酶基因。结论:揭示了参与北豆根活性成分生物合成的关键基因,为进一步研究活性成分代谢途径和调控机制奠定基础。Objective:To analyze the transcriptome information features of Rhizoma menispermi.Methods:The transcriptome of Rhizoma menispermi was sequenced by using second-generation high-throughput sequencing technology,and systematic bioinformatics analysis was performed.Results:Transcriptome sequencing yielded 37,346 Unigenes,with an average length of 986 nt,of which 20,562 Unigenes were annotated.9948 SSRs were identified from 81 kinds of repeat units by MISA method.21 Unigenes may be involved in the biosynthesis of active components of Rhizoma menispermi,involving 8 enzyme genes,including(S)-coclaurine N-methyltransferase,polyphenol oxidase,tyrosine aminotransferase,primary-amine oxidase,aspartate aminotransferase(cytoplasmic),aspartate aminotransferase(mitochondrial),aspartate aminotransferase(chloroplastic)and tyrosine decarboxylase.Conclusion:This data analysis reveals key genes involved in the biosynthesis of the active ingredients of Rhizoma menispermi,laying the foundation for further studies on the metabolic pathways and regulatory mechanisms of the active ingredients.
