摘 要:本研究以大豆浓缩磷脂为原料,从生产中将卵磷脂(PC)副产物磷脂酰乙醇胺(PE)进行纯化,并在超临界CO_(2)条件下进行氢化,后与聚乙二醇单甲醚2000酰氯培化,得到稳定性高的甲氧基聚乙二醇2000-氢化大豆磷脂酰乙醇胺(MPEG2000-HSPE)。采用95%的乙醇,在料液比为1∶3,温度为-13℃的条件下去除副产物中的残余PC,再用90%的石油醚,在料液比为1∶4,温度为30℃的条件下进行萃取,去除肌醇等其他磷脂,使PE纯度达到91.28%,得率为86.83%。在总压力为11 MPa(CO_(2)分压7 MPa、氢气分压4 MPa),Pd/c催化剂用量为4%,氢化温度为60℃,氢化时间为120 min,搅拌速度为300 r/min时,碘值和反式脂肪酸含量显著降低。当氢化磷脂酰乙醇胺(HSPE)与聚乙二醇单甲醚2000酰氯比例为3.5∶1,三乙胺添加量为3 mL时,反应温度50℃,反应时间4 h,产物转化率为84.63%。通过红外光谱分析发现,发现原料中的—NH_(2)消失,产物中出现—NH基团,产物吸光度为0.631、碘值为23.27 gI/100 g、水-正己烷两相分开10 mL时间为392 s,所得产品分散性好、稳定性高。In this study,concentrated soybean phospholipids were used as the raw materials,lecithin(PC)by-product phosphatidylethanolamine(PE)was purified from the production,and hydrogenated under supercritical CO_(2)conditions,and then combined with polyethylene glycol monomethyl ether 2000 acid chloride cultured to obtain high stability methoxypolyethylene glycol 2000-hydrogenated soybean phosphatidylethanolamine(MPEG2000-HSPE).The 95%ethanol was used to remove residual PC from the by-products at a material-to-liquid ratio of 1∶3 and a temperature of-13℃,and then,90%petroleum ether was used to extract under the conditions of a solid-liquid ratio of 1∶4 and a temperature of 30℃to remove other phospholipids,such as inositol,so that the purity of PE reached 91.28%and the yield was 86.83%.When the total pressure was 11 MPa(CO_(2)partial pressure of 7MPa and hydrogen partial pressure of 4 MPa),the amount of Pd/c catalyst was 4%,the hydrogenation temperature was 60℃,the hydrogenation time was 120 min,and the stirring speed was 300 r/min,the iodine value and trans fatty acid content was significantly reduced.When the ratio of hydrogenated phosphatidylethanolamine(HSPE)to polyethylene glycol monomethyl ether 2000 acid chloride was 3.5∶1 and the amount of triethylamine added was 3 mL,the reaction temperature was 50℃,the reaction time was 4 h,and the product conversion rate was 84.63%.Through infrared spectrum analysis,it was found that,—NH_(2) in the raw material disappeared,and—NH group appeared in the product.The absorbance of the product was 0.631,the iodine value was 23.27 gI/100 g,and the separation time of water and n-hexane was 10 mL for 392 s.The product had a good dispersion and high stability.
